摘  要:为探究干酪乳杆菌(Lactobacillus casei)对LPS诱导的奶牛乳腺上皮细胞(BMEC)炎性损伤的缓解作用及机制,将10^(4),10^(5),10^(6)CFU/mL的干酪乳杆菌与奶牛乳腺上皮细胞共培养3h后加入1 mg/L的LPS继续培养8 h.使用荧光定量PCR和Western blot方法检测相关炎性因子mRNA和通路关键蛋白的表达量。结果显示,与对照组相比,10^(5),10^(6)CFU/mL的干酪乳杆菌预处理组显著降低了奶牛乳腺上皮细胞IL-1β、IL-6、IL-8和TNF-αmRNA的表达(P<0.05),同时,10^(4),10^(5),10^(6)CFU/mL的干酪乳杆菌预处理显著降低了IкBα和p65蛋白的磷酸化水平(P<0.05)。结果表明,干酪乳杆菌可以通过抑制奶牛乳腺上皮细胞NF-κB信号通路及相关炎性因子基因的表达,从而发挥抗炎作用。In order to investigate the alleviating effect and mechanism of Lactobacillus casei on LPS-induced inflammatory injury in bovine mammary epithelial cells,Lactobacillus casei with con-centrations of 10^(4),10^(5)and 10^(6)CFU/mL were pretreated with bovine mammary epithelial cells for 3 h,and then 1 mg/L LPS was added,then cells were cultured for 8 h.The expression levels of re-lated inflammatory factors mRNA and key signaling pathway proteins were detected by real time PCR and Western blot.The results showed that compared with the control group,the expressions of IL-1β,IL-6,IL-8 and TNF-α mRNA in bovine mammary epithelial cells reduced significantly in the 10^(5)and 10^(6)CFU/mL Lactobacillus casei pret reatment groups(P<0.05).At the same time,the phosphorylation levels of IкBα and p65 reduced significantly in the pretreatment of Lactobacil-lus casei at 10^(4),10^(5)and 10^(6)CFU/mL(P<0.05).The above results indicated that Lactobacillus casei can exert anti-inflammatory effects by inhibi ting the expression of NF-B signaling pathway and related inflammatory genes in bovine mammary epithelial cells.
