摘  要:目的 应用拉曼光谱技术对单个卵裂胚培养液滴中的胚胎代谢物进行测定分析,寻找一种新的客观评价胚胎质量的方法,优选具有发育潜力的胚胎。方法 收集2019年86例体外受精/卵细胞浆内单精子注射周期中第3天(D3)卵裂期胚胎培养液,其中D3优质卵裂胚形成优质囊胚27例,D3优质卵裂胚未形成囊胚29例,D3次级卵裂胚未形成囊胚30例。通过拉曼光谱系统收集卵裂胚培养液的拉曼光谱,采用主成分分析算法降低数据维度,提取光谱特征,并结合样本类别值建立可实现分类评估的统计模型。结果 优质卵裂胚与次级卵裂胚之间的信号存在明显差异,具有明确的聚类现象,分类准确性可达到85%以上;形成囊胚的卵裂期胚胎和未形成囊胚的卵裂期胚胎之间的信号无明显差异,分类准确性只有57%。结论 发育至D3的优质卵裂胚和次级卵裂胚存在代谢差异,拉曼光谱可区分D3优质卵裂胚和D3次级卵裂胚,但未能有效区分优质卵裂胚能否形成囊胚。Objective Using the Raman spectroscopy to determine and analyze of embryonic metabolites in single cleavage embryo culture droplets,in order to find a new objective method of evaluating the quality of embryos and to select embryos with developmental potential.Methods The culture medium of Day3(D3) cleavage embryos from 86 IVF/ICSI cycles in 2019 was collected:27 cases of D3 high quality cleavage embryos forming high quality blastocysts,29 cases of D3 high quality cleavage embryos not forming blastocysts,30 cases of D3 secondary cleavage embryos not forming blastocysts.The Raman spectra of the culture medium of cleavage embryos were collected by the Raman spectroscopy system,and principal component analysis algorithm was used to reduce the data dimension,extract spectral features,and establish a statistical model for classification evaluation based on the sample category values.Results There was a significant difference in the signal between high quality cleavage embryos and secondary cleavage embryos,with a clear clustering phenomenon and a classification accuracy of more than 85%.There was no significant difference in the signal between blastocyst cleavage embryos and non-blastocyst cleavage embryos,with a classification accuracy of 57%.Conclusion There are metabolic differences between high quality and secondary cleavage embryos developed to D3.Raman spectroscopy can completely distinguish D3 high-quality cleavage embryos from D3 secondary cleavage embryos,and this difference is not significantly related to blastocyst formation ability.
