一例猪蓝耳病与猪圆环病毒病及多重细菌共感染的实验室诊断

摘　　要：为了确诊2021年10月在福建龙岩地区某规模猪场发生的一起仔猪体温升高、呼吸困难、消瘦的病例,分别设计PRRSV、PCV2特异性扩增引物进行PCR检测及阳性样品基因克隆与序列分析。结果表明,送检病例检出PRRSV和PCV2型,PRRSV ORF5序列分析发现所得毒株为高致病性毒株,PCV2全长基因的序列分析为PCV2a亚型;同时从肺脏和肝脏分离所得细菌的16S rRNA进行鉴定与测序,结果发现分离菌为猪链球菌、肺炎克雷伯菌、O157:H7血清型大肠杆菌。说明该猪场疫情主要由高致性PRRSV与PCV2共感染,同时并发多重细菌感染引起。通过对该病例的诊断和疫情分析提出针对性防控措施,为养猪生产实践中的疫病防控提供参考。In order to diagnose a case of piglets’ temperature rises、breathing difficulties and weight loss in a large-scale pig farm in Longyan area, Fujian province in October 2021, PRRSV and PCV2 specific amplification primers were designed respectively, the positive samples were detected by PCR and gene cloning and sequence analysis, the results showed that the PRRSV and PCV2 were detected in the submitted cases, PRRSV ORF5 sequence analysis showed that the obtained strain was highly pathogenic, the sequence analysis of the full-length gene of PCV2 was PCV2a subtype. At the same time, the identification and sequencing results of 16S rRNA of bacteria isolated from lung and liver showed that the isolated bacteria were Streptococcus suis, Klebsiella pneumoniae and O157:H7serotype Escherichia coli. It shows that the epidemic situation in pig farms was mainly caused by the mixed infection of highly pathogenic PRRSV and PCV2, followed by multiple bacterial infections. Through the detection and epidemic analysis of the case, the prevention and control measures are put forward, which provides a reference for prevention and control in production practice.
