摘  要:为了简单、快速、准确地检测水果及其制品中的克百威农药残留,基于单克隆抗体,建立了间接竞争生物素/亲和素酶联免疫分析方法(BA-ELISA).以苹果、苹果汁、橙子、橙汁为样品原材料,采用乙腈提取、旋蒸、甲醇复溶等简单前处理,用0.5%的FG-PBS稀释20倍以基本消除基质影响,用BA-ELISA法测定样品中的克百威含量,同时优化检测条件并对该方法进行评价.结果表明,BA-ELISA检测方法的标准曲线检测限为(0.082±0.002)μg/L,灵敏度为(0.265±0.010)μg/L. 4种水果及其制品中的克百威在3种不同加标浓度水平下的添加回收率为90.20%~99.83%,变异系数为1.8%~6.3%,检测时间缩短至1 h.以气相色谱法作为对照,BA-ELISA法对克百威含量的测定较为准确,可作为水果中检测克百威残留的技术手段.To detect carbofuran pesticide residues in fruit and their products simply, quickly and accurately, an indirect competitive biotin/avidin-enzyme-linked immunosorbent assay(BA-ELISA) was established based on the monoclonal antibody. Apple, apple juice, orange and orange juice were used as raw materials. Simple pretreatment including acetonitrile extraction, rotary steaming and methanol re-dissolution was performed. The influence of the matrix was eliminated by diluting 20 times with 0.5% FG-PBS. The carbofuran content was determined by BA-ELISA. At the same time, the detection conditions were optimized and the method was evaluated. The results showed that the detection limit of the BA-ELISA method is(0.082±0.002)μg/L and the sensitivity is(0.265±0.010)μg/L. The recoveries of carbofuran in the four kinds of fruits and their products were 90.20%-99.83%,the coefficient of variation was 1.8%-6.3%,and the detection time was shortened to 1 h. With the GC method as the control,the BA-ELISA method is proved to be accurate in the determination of carbofuran content. So the BA-ELISA method can be used as a technical means to detect carbofuran residues in fruits.
