摘  要:近年来,在河南省温县地区的中草药种植基地出现了大面积地黄叶部的轮纹病。发病初期叶片表面出现褐色病斑,圆形或近圆形,具有明显同心轮纹,后期病斑易穿孔,最终多个病斑扩散至整张叶片,导致叶片枯死。采集发病期的地黄叶片,分离提取菌株DH20-2基因组DNA,采用CTAB法,对菌株的ITS序列进行PCR扩增。通过系统发育进化树分析,菌株与Fusarium oxysporum聚集在同一分支上;结合柯赫氏法则验证,河南温县地黄轮纹病病原初步确定为尖孢镰刀菌F.o xysporum,研究发现温县地区地黄轮纹病的病原菌范围扩大,为病害的有效防治提供新的依据与思路。In recent years,a large area of leaf ring rot of Rehmannia glutinosa Libosch has been found in Chinese herbal medicine planting bases in County Wen,Henan Province,China.Brown lesions appeared on the surface of leaves at the beginning of the disease,round or nearly round,with obvious concentric ring spots,and the lesions were easy to perforate in the later stages.At the end,multiple lesions spread to the entire leaf,causing the death of leaf.The leaves of rehmannia glutinosa during the disease stage were collected in order to isolate genomic DNA of strain DH20-2,then ITS sequence of strain DH20-2was amplified by PCR,adopting CTAB method.Phylogenetic tree analysis showed that strain DH20-2and Fusarium oxysporum clustered on the same branch.Based on Koch’s postulates,it was verified that this pathogen of rehmannia in County Wen was F.oxysporum.This study expanded the pathogen research of rehmannia in County Wen and provided new basis and ideas for effective control of this disease.
