摘  要:通过计算机技术筛选柴胡、白芍治疗抑郁症的主要活性成分及其潜在作用靶点,探讨其抗抑郁症的分子机制.应用TCMSP数据库总结整理柴胡白芍的成分和作用靶点,以OB≥30%、DL≥0.18为标准筛选出有效成分,运用Uniprot数据库查询靶蛋白对应的基因名.通过数据库提取MDD疾病的相关靶点基因,进行Venn分析选取两者交集的靶蛋白,导入String数据库进行蛋白质相互作用分析,cytoscape3.8.0软件建立成分-靶点-疾病网络进行拓朴异构分析,再采用DAVID在线分析数据工具和FunRich对筛选出的共同靶点进行GO和KEGG富集分析.筛选得到29个活性成分,与52个抗抑郁的靶点相关等;PPI网络涉及52个节点,367条边;以P<0.05进行筛选,富集得到GO生物过程236条,主要涉及对雌二醇反应、信号转导、细胞外空间、与类固醇结合等.富集KEGG通路67条,主要为神经活性配体-受体相互作用等相关通路、癌症通路、雌激素信号通路等.探究了柴胡白芍药对治疗抑郁症的活性成分、作用靶点及相关通路,为柴胡白芍治疗抑郁症的后续研究提供了理论依据.To screen the main active components of Bupleurum and Paeonia lactiflora in the treatment of depression and their potential targets by computer technology,and to explore the molecular mechanism of their antidepressant.The TCMSP database was used to summarize and sort out the components and targets of Bupleurum Radix Paeoniae Alba,and the active ingredients were screened out with OB≥30%and DL≥0.18.The Uniprot database was used to query the gene names corresponding to the target proteins.Therelevant target genes of MDD diseases were extracted from the database,and Venn analysis was performed to select the target proteins of the intersection of the two,and imported into the String database for protein interaction analysis.GO and KEGG enrichment analysis was performed on the screened common targets using DAVID online data analysis tool and FunRich.29 active ingredients were screened,which were related to 52 antidepressant targets,etc.;the PPI network involved 52 nodes and 367 edges;P<0.05 was screened,and 236 GO biological processes were enriched,mainly involving the Estradiol response,signal transduction,extracellular space,binding to steroids,etc.There are 67 KEGG pathways enriched,mainly related to neuroactive ligand-receptor interaction pathways,cancer pathways,and estrogen signaling pathways.This paper preliminarily explored the active components,targets and related pathways of Bupleurum chinensis in the treatment of depression,and provided a theoretical basis for the follow-up study of Bupleurum radix in the treatment of depression.
