摘 要:目的:探讨Omega-3脂肪酸(ω-3 PUFA)对金黄地鼠痤疮模型皮脂腺斑、过氧化物酶体增殖物激活受体(PPARγ)及硬脂酰辅酶A去饱和酶(SCD-1)的影响。方法:将24只金黄地鼠随机分为空白对照组、ω-3 PUFA低剂量组、ω-3 PUFA高剂量组及阳性对照组,连续灌胃4周。测量各组金黄地鼠皮脂腺斑面积,观察其组织病理改变。采用免疫组化检测皮脂腺斑组织中PPARγ和SCD-1蛋白表达。结果:干预后,与空白对照组比较,ω-3 PUFA低剂量组、ω-3 PUFA高剂量组及阳性对照组金黄地鼠皮脂腺斑面积均较小,且各组皮脂腺斑PPARγ蛋白表达均升高(P<0.05),而SCD-1蛋白表达均减弱(P<0.05)。结论:ω-3PUFA能够改善金黄地鼠痤疮模型皮脂腺斑大小和组织结构,其机制可能是通过调节PPARγ/SCD-1信号通路。Objective:To investigate the effects of Omega-3 fatty acids(ω-3 PUFA) on sebaceous gland,peroxisome proliferator-activated receptor(PPAR)γ and stearoyl-CoA desaturase 1(SCD-1) in a golden hamster acne model.Methods:Twenty-four golden hamsters were randomly divided into blank control group,ω-3 PUFA low-dose group,ω-3 PUFA high-dose group and positive control group,followed by gavage for 4 weeks.The size of sebaceous glands in each group was measured,histopathological changes were observed,and the protein expression of PPARγ and SCD-1 in sebaceous glands was detected by immunohistochemical method.Results:After intervention,compared with that in the blank control group,the area of sebaceous glands was smaller in other groups.Compared with that in the blank group,the expression of PPARγ protein in sebaceous spots of golden hamster was increased(P<0.05),while the expression of SCD-1 protein was decreased(P<0.05) in other groups.Conclusion:ω-3 PUFA increases sebaceous gland spot size and optimizes tissue structure in the golden hamster acne model,possibly through regulating the PPARγ/SCD-1 signaling pathway.
