摘 要:【目的】探讨苦参素对青光眼模型大鼠的视网膜神经保护作用及机制。【方法】将40只SD大鼠随机分为正常组、模型组、法舒地尔组和苦参素组。除正常组,其余各组大鼠采用532-二极管激光光凝法建立青光眼模型。自模型制备当日起,开始进行药物干预。干预结束后,采用苏木素-伊红(HE)染色法观察大鼠视网膜病理形态,并测量视网膜厚度,采用免疫荧光染色法检测大鼠视网膜组织内Brn3a及Tau、突触素表达,并计数Brn3a阳性标记的视网膜神经节细胞(RGCs)数量,采用蛋白免疫印迹(Western Blot)法检测大鼠视网膜组织磷酸化LIM激酶(p-LIMK)、磷酸化肌球蛋白轻链磷酸酶(pMLCP)、磷酸化肌球蛋白轻链(p-MLC)、Rho激酶(ROCK)1和ROCK2的蛋白表达。【结果】与正常组比较,模型组大鼠视网膜厚度降低,RGCs数量减少(P<0.05);法舒地尔组和苦参素组大鼠视网膜厚度和RGCs数量较模型组升高(P<0.05);法舒地尔组与苦参素组间视网膜厚度及RGCs数量比较,差异均无统计学意义(P>0.05)。模型组大鼠视网膜组织Tau、突触素,p-LIMK、p-MLCP、p-MLC、ROCK1和ROCK2蛋白表达水平均低于正常组(P<0.05);法舒地尔组和苦参素组大鼠视网膜组织内Tau、突触素,p-LIMK、p-MLCP、p-MLC、ROCK1和ROCK2蛋白表达水平均高于模型组(P<0.05);法舒地尔组与苦参素组间各蛋白表达水平无明显差异(P>0.05)。【结论】苦参素可促进青光眼大鼠RGCs的存活和轴突的再生,其机制可能与抑制ROCK的活性有关。Objective To explore the retinal neuroprotective effect of matrine for glaucoma model rats and its mechanism.Methods Forty SD rats were randomly divided into the normal group,the model group,the fasudil group and the matrine group.Glaucoma model was established using 532-diode laser photocoagulation in all groups except the normal group.The drug intervention was started from the day of model preparation.At the end of the intervention,the pathological morphology of the rat retina was observed by hematoxylin-eosin(HE)staining and the retinal thickness was measured.The protein expression of Brn3a and Tau and synaptophysin in the rat retinal tissues was detected by immunofluorescence staining,and the number of Brn3a-positive labeled retinal ganglion cells(RGCs)was counted.The protein expression of phosphorylated LIM kinase(p-LIMK),phosphorylated myosin light chain phosphatase(p-MLCP),phosphorylated myosin light chain(p-MLC),Rho kinase(ROCK)1and ROCK2 in rat retinal tissues was measured by Western Blot.Results Compared with the normal group,the retinal thickness and the number of RGCs in the model group were decreased(P<0.05);the retinal thickness and the number of RGCs in the fasudil group and the matrine group were increased as compared with those in the model group(P<0.05);there was no significant difference in retinal thickness and number of RGCs between the fasudil group and the matrine group(P>0.05).The protein expression levels of Tau,synaptophysin,p-LIMK,pMLCP,p-MLC,ROCK1 and ROCK2 in the model group were lower than those in the normal group(P<0.05);the protein expression levels of Tau,synaptophysin,p-LIMK,p-MLCP,p-MLC,ROCK1 and ROCK2 in the fasudil group and the matrine group were higher than those in the model group,the differences being statistically significant(P<0.05);there were no significant differences in protein expression levels of the above indexes between the fasudil group and the matrine group(P>0.05).Conclusion Matrine promotes the survival of RGCs and axonal regeneration in glaucomatous rats,and
