摘 要:【目的】观察补肾健脾方对db/db小鼠糖代谢及骨代谢指标的影响。【方法】将16只雄性db/db阳性小鼠随机分为治疗组和阳性对照组各8只,8只BKS背景阴性小鼠随机分为阴性对照组1和阴性对照组2各4只。治疗组、阴性对照组1给予补肾健脾方冻干粉溶液灌胃,阳性对照组、阴性对照组2给予等体积生理盐水灌胃。给药4周后,断颈处死,腹主动脉取血,检测小鼠血清脂联素(APN)、25-羟基维生素D3[25-(OH)D3]、Ⅰ型前胶原氨基端原肽(PINP)、血糖、胰岛素等指标,苏木素-伊红(HE)染色观察胰岛病理学变化,末端脱氧核苷酸转移酶介导的dUTP缺口末端标记(TUNEL)法观察胰岛细胞凋亡情况,免疫组织化学法观察小鼠胰腺凋亡相关蛋白Bax、Bcl-2、Caspase-3阳性表达情况。【结果】治疗4周后:治疗组小鼠胰岛TUNEL染色面积百分比低于阳性对照组(P<0.01);治疗组小鼠血清APN、25-(OH)D3、PINP水平明显高于阳性对照组(P<0.01);治疗组小鼠空腹血糖、空腹胰岛素水平低于阳性对照组(P<0.01);与阳性对照组比较,治疗组小鼠胰腺Bax、Caspase-3阳性面积百分比降低(P<0.05或P<0.01),Bcl-2阳性面积百分比升高(P<0.01)。【结论】补肾健脾方具有调节db/db小鼠糖代谢及骨代谢的作用,其机制可能与抑制胰岛细胞凋亡或直接调节糖-骨代谢指标有关。Objective To observe the effects of Bushen Jianpi Formula on glucose metabolism and bone metabolic indexes in db/db mice.Methods Sixteen male db/db positive mice were randomly divided into the treatment group and the positive control group,with 8 mice in each group,while 8 BKS background negative mice were randomly divided into the negative control group 1 and negative control group 2,with 4 mice in each group.The treatment group and negative control group 1 were given lyophilized powder solution of Bushen Jianpi Formula by gavage,while the positive control group and negative control group 2 were given equal volume of normal saline by gavage.After 4 weeks of administration,the mice were executed by neck amputation,and blood was collected from the abdominal aorta to detect the serum adiponectin(APN),25-hydroxyvitamin D3[25-(OH)D3],aminoterminal propeptide of typeⅠprocollagen(PINP),blood glucose and insulin,morphological changes of pancreas islet were observed by hematoxylin-eosin(HE)staining,apoptosis of islet cells was observed by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling(TUNEL)method,positive expression of Bax,Bcl-2 and Caspase-3 protiens related to apoptosis in mouse pancreas was observed by immunohistochemistry.Results After 4 weeks of treatment:the percentage of TUNEL-stained area of pancrea of mice in the treatment group was lower than that of the positive control group(P<0.01);the serum APN,25-(OH)D3 and PINP levels of mice in the treatment group were significantly higher than those of the positive control group(P<0.01);the fasting blood glucose and fasting insulin levels of mice in the treatment group were lower than those of the positive control group(P<0.01);compared to the positive control group,the percentage of pancreatic Bax and Caspase-3 positive area was decreased(P<0.05 or P<0.01),and the percentage of Bcl-2 positive area was increased(P<0.01)in the treatment group.Conclusion Bushen Jianpi Formula has effects of regulating glucose metabolism and lipid metabolism in db
