摘 要:【目的】蒿属植物种类多,分布地域广,种间差异小,形态学等常规方法难以有效鉴定和区分不同种质资源。明晰国内藜蒿不同种质资源间的亲缘关系,保护其遗传多样性。【方法】利用RAPD分子标记,对6个省份共102份藜蒿样品DNA进行PCR扩增,扩增结果用POPGENE32、STRUCTURE等软件分别进行遗传多样性指标计算、聚类分析和群体结构分析。【结果】共有11条RAPD引物扩增出95条带,其中多态性条带93条,多态性比率为97.89%。102份藜蒿资源的种群间基因分化系数(Gst)为0.4109,基因流(Nm)为0.7170,遗传相似系数在0.61~0.98之间。【结论】供试的102份藜蒿资源遗传变异主要发生在群体内,存在一定程度的基因流动,具有丰富的遗传多样性。UPGMA聚类和贝叶斯聚类均将102份藜蒿资源分为3类。【Objective】Artemisia has many species,wide distribution areas,small interspecific differences,and conventional methods such as morphology are difficult to effectively identify and distinguish different germplasm resources.Artemisia selengensis has important economic development prospects.In order to clarify the genetic relationship between different germplasm resources of Artemisia selengensis in China and protect its genetic diversity.【Method】This study used RAPD molecular markers to amplify the DNA of 102 Artemisia selengensis samples from 6 provinces by PCR.The amplification results were calculated by popgene 32,structure and other software for genetic diversity index calculation,cluster analysis and population structure analysis.【Result】A total of 11 RAPD primers amplified 95 bands,including 93 polymorphic bands,with a polymorphism ratio of 97.89%.Among 102 Artemisia selengensis resources,the gene differentiation coefficient GST is 0.4109,the gene flow(nm)is 0.7170,and the genetic similarity coefficient is between 0.61 and 0.98.【Conclusion】The genetic variation of 102 Artemisia selengensis resources tested mainly occurred in the population,with a certain degree of gene flow and rich genetic diversity.Both UPGMA and Bayesian clustering classified 102 A.selengensis resources into 3 categories.
