摘　　要：为了研究葡萄籽原花青素对新疆驴精液低温保存效果的影响,试验在新疆驴精液稀释液中添加0(对照),0.02,0.04,0.06,0.08 g/L的葡萄籽原花青素进行低温保存,采用倒置显微镜检测新疆驴精子活率、质膜完整率、顶体完整率,应用超氧化物歧化酶(SOD)和过氧化氢酶(CAT)测定试剂盒检测SOD和CAT活性,分析不同组别精液品质的差异。结果表明:与对照组相比,添加0.06 g/L葡萄籽原花青素且保存至第7天的精液中精子活率、质膜完整率、顶体完整率显著提高(P<0.05);保存至第5天时,0.02,0.04,0.06,0.08 g/L组的SOD活性显著高于对照组(P<0.05);保存至第7天时,0.04 g/L组的CAT活性最高。说明在稀释液中添加0.06 g/L的葡萄籽原花青素有助于提高新疆驴精液低温保存的质量。To study the effect of grape seed proanthocyanidins on cryopreservation of Xinjiang donkey semen,different concentrations of grape seed proanthocyanidins(0,0.02,0.04,0.06,0.08 g/L)were added into Xinjiang donkey semen diluent and stored at low temperature.The viability,plasma membrane integrity and acrosome integrity of Xinjiang donkey sperm were detected by inverted microscope,and the activities of superoxide dismutase(SOD)and catalase(CAT)were detected by kit.The differences of semen quality in different groups were analyzed.The results showed that compared with the control group,the sperm motility rate,plasma membrane integrity and acrosome integrity in semen supplemented with 0.06 g/L grape seed proanthocyanidins when it was stored until day 7 were significantly increased(P<0.05).The SOD activity in the 0.02,0.04,0.06,0.08 g/L groups was significantly higher than that in the control group when semen was stored until day 5(P<0.05).The CAT activity was the highest in the 0.04 g/L group when semen was stored until day 7(P<0.05).The results indicated that adding 0.06 g/L grape seed proanthocyanidins in the diluent could improve the cryopreserve quality of Xinjiang donkey semen.
