摘 要:目的 运用网络药理学方法探讨宣白承气汤治疗支气管哮喘的作用机制。方法 通过TCMSP数据库筛选宣白承气汤的活性成分及作用靶点,通过GeneCards数据库筛选支气管哮喘的疾病相关靶点,获取二者交集靶点,即宣白承气汤治疗支气管哮喘的潜在作用靶点。使用Cytoscape 3.7.2软件构建“疾病-药物-活性成分-靶点”网络,并分析网络中的核心活性成分;将交集靶点导入STRING数据库进行蛋白互作(PPI)网络分析,筛选出潜在核心靶点;运用R语言软件包对交集靶点进行GO功能和KEGG通路富集分析。结果 共筛选得到34个活性成分及64个潜在作用靶点(交集靶点);获得香叶木素、羟基芫花素、泽兰黄醇素、α-菠菜甾醇等核心活性化合物,IL-6、MAPK3、MMP-9、PPARG和ICAM1等核心靶点;共筛选得到67条信号通路,关键通路包括神经活性配体-受体相互作用信号通路、c AMP信号通路、钙信号通路、TNF信号通路、Th17细胞分化、Th1/Th2细胞分化等。结论 宣白承气汤可能是通过香叶木素、羟基芫花素等关键活性成分,作用于IL-6、MAPK3、MMP-9等潜在核心靶点,调控神经活性配体-受体相互作用、cAMP等信号通路,从而发挥治疗支气管哮喘的作用。Objective To explore the mechanism of action of Xuanbai Chengqi Decoction in the treatment of bronchial asthma(BA)by network pharmacology. Methods The active components and targets of Xuanbai Chengqi Decoction were screened through the TCMSP database,and the disease-related targets of BA were screened through the GeneCards database to obtain the intersecting targets, namely, the potential targets of Xuanbai Chengqi Decoction for the treatment of BA. The "disease-drug-active component-target" network was constructed by Cytoscape 3.7.2 software, and the core active components in the network were analyzed;the intersection targets were imported into the STRING database for protein-protein interaction(PPI)network analysis to screen out potential core targets;the GO function and KEGG pathway enrichment of the intersection targets were analyzed by R language packages. Results A total of 34 active components and 64 potential targets(intersection targets) were screened;core active compounds such as diosmetin, hydroxygenkwanin, eupatin, spinasterol, and core targets such as IL-6, MAPK3, MMP-9, PPARG and ICAM1 were obtained;a total of 67 signalling pathways were screened,including neuroactive ligand-receptor interaction signalling pathway,cAMP signalling pathway,calcium signalling pathway, TNF signalling pathway, Th17 cell differentiation, Th1 and Th2 cell differentiation, etc.Conclusion Xuanbai Chengqi Decoction may act on potential core targets such as IL-6, MAPK3 and MMP-9through key active components such as diosmetin and hydroxygenkwanin,and modulate signalling pathways such as neuroactive ligand-receptor interactions and cAMP to exert therapeutic effects on BA.
