摘 要:目的 探讨炆制对何首乌中蒽醌糖苷类成分的影响及其减毒效应。方法 采用液相色谱-质谱联用(LC-MS)法对何首乌药材及其不同炮制品的蒽醌糖苷类成分进行定性分析;进一步采用正常人肝细胞系(L02肝细胞)作为评价模型,以谷丙转移酶(ALT)、谷草转移酶(AST)、谷氨酰胺转肽酶(GGT)、乳酸脱氢酶(LDH)为评价指标,对何首乌不同炮制品及变化的蒽醌糖苷类成分的肝脏毒性进行评价。结果 何首乌炮制前后的变化成分为蒽醌糖苷类成分大黄素-8-O-β-D-葡萄糖苷和大黄素甲醚-8-O-β-D-葡萄糖苷。肝细胞毒性实验中,何首乌不同炮制品毒性由大到小依次为:何首乌药材>制何首乌饮片>九蒸九晒何首乌饮片>炆何首乌饮片;炆制后消失的大黄素-8-O-β-D-葡萄糖苷和大黄素甲醚-8-O-β-D-葡萄糖苷具有肝脏毒性,且随着浓度的减小,肝毒性降低。结论 何首乌的多种炮制方法均有减毒作用,以炆制为优;炆制后消失的蒽醌糖苷类成分被证实具有肝脏毒性,推测蒽醌糖苷类成分为何首乌肝脏毒性的物质基础,为临床安全用药提供了依据。Objective To investigate the effect of simmering Polygonum multiflorum Radix on its anthraquinone glycosides and the attenuation effect. Methods Liquid chromatography-mass spectrometry(LC-MS) method was used to qualitatively analyze the anthraquinone glycosides in Polygonum multiflorum Radix and its different processed products. Furthermore,normal human hepatocytes L02 were used as evaluation model,and ALT,AST,GGT and LDH of L02 cells were used as evaluation indexes for the hepatotoxicity of different processed products of Polygonum multiflorum Radix and the changed anthraquinone glycosides. Results The changes of anthraquinone glycosides were emodin-8-O-β-D-glucoside and physcion-8-O-β-D-glucoside before and after processing of Polygonum multiflorum Radix. Poignant In hepatocyte toxicity test, the order of toxicities of different processed products of Polygonum multiflorum Radix is as follows: Polygonum multiflorum Radix>prepared Polygonum multiflorum Radix slices>nine-time repeat of the steaming and sun-drying of Polygonum multiflorum Radix slices>simmered Polygonum multiflorum Radix slices. The emodin-8-O-β-D-glucoside and physcion-8-O-β-D-glucoside which disappeared after simmering also have hepatotoxicity,and the hepatotoxicity decrease with their decreasing concentrations. Conclusion Various processing methods of Polygonum multiflorum Radix have detoxification effect,and simmering showed the best detoxification effect. The anthraquinone glycosides that disappeared after simmering were proved to have hepatotoxicity. It is speculated that the anthraquinone glycosides are the material basis of the hepatotoxicity of Polygonum multiflorum Radix,which provides basis for clinical safe drug use.
