摘 要:为了观察酸碱溶液、氯化钠和饮酒对尿酸酶缺失大鼠(昆明-DY大鼠)尿酸血的影响.将45 d日龄雄性昆明-DY大鼠自由饮用醋酸氨丁三醇(0.5和1.5 g/L,pH8.4)、盐酸氨丁三醇(0.5和1.5 g/L,pH8.4)、碳酸氢钠(0.5和1.5g/L,pH8.4)、醋酸钠(0.5和1.5 g/L,pH8.4)、氯化钠(0.5和1.5 g/L,pH6.5)、醋酸(0.5和1.5 g/L,pH4.0)、盐酸(0.5 g/L,pH2.0)、乙醇(4 mL/L)溶液和弱碱性矿泉水(pH9.3)14 d,实验前后断尾取血制备血清,收集大鼠24 h尿液和粪便,记录24 h进食和饮水量.样品中的尿酸含量采用磷钨酸法检测.结果发现,醋酸氨丁三醇、盐酸氨丁三醇、碳酸氢钠、氯化钠和盐酸能降低大鼠血清尿酸,乙醇能升高血尿酸,醋酸钠、醋酸和弱碱性矿泉水对大鼠血尿酸无明显影响.综上,氨丁三醇醋酸盐溶液在口服降尿酸方面具有一定优势,禁酒对阻止尿酸升高有利,而弱碱性矿泉水降尿酸作用不明显.To observe serum uric acid(SUA) in Kunming-DY rats(an uricase-deficient rats) affected by sodium chloride solutions, alcoholand other acid and alkaline solutions, male Kunming-DY rats at the age of 45 days were arranged to freely drink tromethamine acetate solutions(0.5 or 1.5 g/L, pH8.4), tromethamine hydrochloride solutions(0.5 or 1.5 g/L,pH8.4), sodium bicarbonate solution(0.5 or 1.5 g/L, pH8.4), sodium acetate solutions(0.5 or 1.5 g/L, pH8.4), sodium chloride solutions(0.5 or 1.5 g/L,pH6.5), acetic acid solutions(0.5 or 1.5 g/L, pH4.0), hydrochloride acid solution(0.5 g/L,pH2.0), alcohol(4 mL/L) and weakly alkaline mineral water(pH 9.3) for 14 d. Blood samples were collected by cutting their tails before and after the experiment, and serum was obtained by spinning. The urine and stool they excreted during 24 h were collected, and the food and water(or solution) they consumed during 24 h were recorded before and after the experiment, too. Uric acid in the samples were determined using uric acid assay kits of phosphotungstic acid method. Results showed that SUA was lowered by the solutions of tromethamine acetate, tromethamine hydrochloride, sodium bicarbonate, sodium chloride, or hydrochlorideand was increased by alcohol solution. It can be found that SUAwas almost not affected by sodium acetate solutions and the weakly alkaline mineral water. Thus, oral tromethamine acetate has the advantage to lower SUA. Prohibition of alcohol is helpful to prevent the increase of uric acid, and the SUA-lowering effect of weakly alkaline mineral water is too weak to mention.
