摘 要:【目的】在细胞水平探讨减味二藤汤(红藤、忍冬藤等组成)对宫腔机械性损伤所致宫腔粘连的治疗作用机制。【方法】对宫腔机械性损伤所致宫腔粘连大鼠及正常大鼠的子宫内膜基质细胞(ESCs)进行体外原代培养,所培养的细胞进行免疫荧光染色法鉴定,共分为正常组、模型组、减味二藤汤含药血清组、空白血清组、拮抗剂组[减味二藤汤含药血清+过氧化物酶体增殖物激活受体α(PPAR-α)抑制剂GW6471],其中,正常组为正常大鼠来源的ESCs,其余各组为宫腔粘连大鼠来源的ESCs。各组细胞处理48 h后,采用细胞计数试剂盒8(CCK-8)法观察细胞活性,采用Western Blot法检测各组细胞PPAR-α、酰基辅酶A氧化酶1(ACOX1)、肉毒碱棕榈酰基转移酶1A(CPT1A)、α-平滑肌肌动蛋白(α-SMA)、Ⅰ型胶原蛋白(CollagenⅠ)及纤连蛋白(Fibronectin)的表达。【结果】与空白血清组和拮抗剂组比较,减味二藤汤含药血清组细胞活性下降,α-SMA、Collagen I、Fibronectin的蛋白表达水平降低(P<0.05),PPAR-α、ACOX1、CPT1A的蛋白表达水平升高(P<0.05)。【结论】减味二藤汤可激活PPAR-α/脂肪酸氧化(FAO)通路,促进脂肪酸氧化,从而改善宫腔机械性损伤引起的宫腔粘连纤维化。Objective To explore the mechanism of action of modified Erteng Decoction(MED,composed of Gaulis Sargentodoxae and Caulis Loniceraejaponicae,and so on)in the treatment of intrauterine adhesion(IUA)induced by mechanical injury of the uterine cavity at the cell level.Methods Endometrial stromal cells(ESCs)of rats with mechanical injury rats and normal rats were cultured primarily in vitro,and the cultured cells were identified by immunofluorescence staining.The ESCs were divided into the normal group,model group,MEDcontaining serum group,blank serum group and antagonist group[modified Erteng Decoction-containing serum+peroxisome proliferator activator receptorα(PPAR-α)inhibitor GW6471],in which the normal group was ESCs from normal rats and the other groups were ESCs from IUA rats.After 48 hours of treatment in each group of ESCs,the cellular activity was observed by cell count kit 8(CCK-8),and the expression of PPAR-α,acyl coenzyme A oxidase 1(ACOX1),carnitine palmitoyltransferase 1A(CPT1A),α-smooth muscle actin(α-SMA),collagen typeⅠ(CollagenⅠ)and Fibronectin in ESCs were detected by Western Blot.Results Compared with the blank serum group and antagonist group,the cellular activity of MED-containing serum group was decreased,the protein expression levels ofα-SMA,CollagenⅠand Fibronectin were decreased(P<0.05),and the protein expression levels of PPAR-α,ACOX1 and CPT1A were increased(P<0.05).Conclusion MED activates the PPAR-α/FAO signaling pathway and promotes fatty acid oxidation,thereby improving fibrosis caused by IUA.
