摘  要:目的:探究超微加味丹参饮对氯化钴(CoCl_(2))诱导损伤的大鼠冠状动脉血管内皮细胞单核细胞趋化蛋白-1(MCP-1)和血管内皮细胞钙黏蛋白(VE-cadherin)表达的影响。方法:取20只大鼠采用随机数字表法分为空白血清组、超微加味丹参饮高剂量组、超微加味丹参饮中剂量组、超微加味丹参饮低剂量组,每组5只。各给药组大鼠灌胃给予相应药物,空白血清组大鼠灌胃给予等体积的蒸馏水,1次/d,连续给药7 d。行腹主动脉采血制备含药血清和空白血清。体外培养大鼠冠状动脉内皮细胞,应用CoCl_(2)诱导建立内皮细胞损伤模型,检测对照组、模型组、超微加味丹参饮高剂量组、超微加味丹参饮中剂量组、超微加味丹参饮低剂量组内皮细胞的细胞生存率、氧化应激指标、凋亡相关蛋白及MCP-1、VE-cadherin的表达水平。结果:与对照组比较,模型组内皮细胞的细胞生存率、SOD活性及Bcl-2蛋白相对表达量均明显降低(P<0.05),MDA和ROS含量、Bax和cleaved Caspase-3蛋白相对表达量及MCP-1和VE-cadherin表达水平均明显升高(P<0.05);与模型组比较,超微加味丹参饮高、中、低剂量组内皮细胞的细胞生存率、SOD活性及Bcl-2蛋白相对表达量均明显升高(P<0.05),MDA和ROS含量、Bax和cleaved Caspase-3蛋白相对表达量及MCP-1和VE-cadherin表达水平均明显降低(P<0.05),且超微加味丹参饮高、中、低剂量组间内皮细胞中上述指标比较,差异均有统计学意义(P<0.05)。结论:超微加味丹参饮对CoCl_(2)诱导损伤的大鼠冠状动脉内皮细胞具有良好的保护作用,可有效降低MCP-1和VE-cadherin的表达。Objective:To investigate the effect of Ultramicro Jiawei Danshen Yin on expression of monocyte chemoattractant protein-1(MCP-1)and vascular endothelial cadherin(VE-cadherin)of injured rats'coronary artery endothelial cells induced by cobalt chloride(CoCl_(2)).Methods:20 rats were randomly divided into blank serum group,Ultramicro Jiawei Danshen Yin high,medium and low dose group with 5 rats in each group.Rats in each administration group were given corresponding drugs by gavage,and rats in blank serum group were given equal volume of distilled water by gavage,once a day,for 7 consecutive days.Blood was collected from abdominal aorta to prepare drug containing serum and blank serum.The rats'coronary artery endothelial cells were cultured in vitro,and the endothelial cell injury model was induced by CoCl_(2).The cell survival rate,oxidative stress index,expression levels of apoptosis related proteins,MCP-1,and VE-cadherin in the control group,model group,high,middle and low dose group of Ultramicro Jiawei Danshen Yin were detected.Results:Compared with the control group,the cell survival rate,SOD activity and relative expression of Bcl-2 protein of endothelial cells in the model group were significantly reduced(P<0.05),and the contents of MDA and ROS,the relative expression of Bax and cleaved Caspase-3,and the expression levels of MCP-1 and VE-cadherin were significantly ncreased(P<0.05).Compared with the model group,the cell survival rate,SOD activity and relative expression of Bcl-2 protein of endothelial cells in the high,medium and low dose groups of Ultramicro Jiawei Danshen Yin were significantly higher(P<0.05),and the contents of MDA and ROS,the relative expression of Bax and cleaved Caspase-3 protein,and the expression levels of MCP-1 and VE-cadherin in endothelial cells in the high,medium and low dose groups of Ultramicro Jiawei Danshen Yin were significantly lower(P<0.05),The difference was statistically significant(P<0.05).Conclusion:Ultramicro Jiawei Danshen Yin had a good protective effect on injured c
