摘 要:[目的]探讨藻蓝蛋白(C-PC)联合光动力疗法对SW1990胰腺癌细胞的抑制作用,并从免疫调节方面初步探讨其作用机制。[方法]实验分成6组:正常组、模型组、CY组、激光照射组、藻蓝蛋白组(C-PC组)、藻蓝蛋白+激光照射组(C-PC+激光照射组)。除正常组外,余组将SW1990细胞接种于小鼠右前肢腋窝处构建小鼠异体移植瘤模型。造模成功后测量肿瘤直径,称量脾重,计算脾指数、肿瘤体积及抑瘤率。采用ELISA法测定小鼠脾细胞培养液和血清中TNF-α的水平以及血清中IL-6的水平。[结果]与模型组比较,C-PC组瘤重显著减少(P<0.05);CY组、C-PC+激光照射组瘤重和瘤体积均显著减少(P<0.01和P<0.05);激光照射组对瘤重和瘤体积无显著影响。激光照射组、C-PC组、C-PC+激光照射组的抑瘤率分别为(39.42±8.47)%、(42.85±12.15)%、(74.80±11.85)%,而C-PC+激光照射组的抑瘤率与CY组[(78.58±8.87)%]相当。与正常组比较,模型组的脾重和脾指数显著减少(P<0.05);与模型组比较,激光照射组、C-PC组和C-PC+激光照射组均能显著增加脾重和脾脏指数(P<0.05或P<0.01),而CY组小鼠脾重和脾指数显著下降(P<0.05)。与正常组比较,模型组TNF-α及IL-6水平无显著性差异(P>0.05);与模型组比较,C-PC组、C-PC+激光照射组均能显著增加小鼠TNF-α及IL-6的水平(P<0.05或P<0.01);而CY组TNF-α及IL-6的水平显著下降(P<0.05或P<0.01);激光照射组可促进IL-6含量(P<0.05),而对TNF-α水平无显著影响(P>0.05)。[结论]藻蓝蛋白联合光动力疗法能有效抑制肿瘤的生长,提高患癌小鼠免疫器官脾脏的免疫功能,增强患癌小鼠血清中TNF-α和IL-6的表达以及脾细胞培养液上清中TNF-α的表达水平,增强机体的体液免疫功能,提示藻蓝蛋白联合光动力疗法可能通过调节机体的免疫功能来达到抑制肿瘤生长的目的。[Objective]To investigate the antitumor effect of C-phycocyanin mediated photodynamic therapy on mice inoculated with SW1990 pancreatic cancer cells,and to investigate its mechanism from the aspect of immune regu⁃lation.[Methods]The experiment was divided into 6 groups:normal group,model group,CY group,laser irradiation group,C-phycocyanin group(C-PC group),C-phycocyanin+laser irradiation group(C-PC+laser group).Except for the normal group,the other groups inoculated SW1990 cells into the axilla of the right forelimb of mice to construct a mouse xenograft tumor model,the tumor diameter,spleen weigh,spleen index,tumor volume and tumor inhibition rate were measured after successful modeling.The expression of TNF-αand IL-6 in spleen cell culture medium and serum of mice were determined by ELISA.[Results]Compared with the model group,the tumor weight in the C-PC group was significantly decreased(P<0.05);the tumor weight and tumor volume in the CY group and the C-PC+laser irradiation group were significantly decreased(P<0.01 and P<0.05).Tumor weight and tumor volume were not significantly affect⁃ed in the laser irradiation group.The tumor inhibition rates of the laser irradiation group,C-PC group,and C-PC+laser irradiation group were(39.42±8.47)%,(42.85±12.15)%,and(74.80±11.85)%,respectively.The tumor inhibition rate of C-CP+laser group was comparable to the CY group[(78.58±8.87)%].Compared with the normal group,the spleen weight and spleen index of the model group were significantly decreased(P<0.05);compared with the model group,the spleen weight and spleen index of the laser irradiation group,C-PC group and C-PC+laser irradiation group could sig⁃nificantly increase(P<0.05 or P<0.01),while the spleen weight and spleen index of mice in the CY group decreased significantly(P<0.05).Compared with the normal group,there was no significant difference in the levels of TNF-αand IL-6 in the model group(P>0.05);compared with the model group,the C-PC group and C-PC+laser irradiation group could significantly increa
