摘 要:为了探究SABATH甲基转移酶家族成员在水稻中的功能,以粳稻品种中花11为实验材料,采用CRISPR-Cas9基因编辑技术对该家族成员OsMT1进行基因编辑,通过测序分析获得了5种编辑类型的突变体,包括3种纯合编辑突变体和2种杂合编辑突变体.分别用50、100和150 mmol/L的NaCl溶液对获得的OsMT1纯合突变体进行盐胁迫处理实验,结果表明,OsMT1突变体的苗长及根长明显长于野生型的苗长及根长,说明OsMT1突变体降低了水稻对盐胁迫的敏感性,增强了水稻对盐胁迫的抗性.To reveal the function of SABATH methyltransferases family members in rice,Japonica Rice Variety Zhonghua 11 was taken as experimental material and the editing mutants of OsMT1 gene was created using CRISPR-Cas9 gene editing technique.Five types of editing mutants including three homozygous editing mutants and two heterozygous editing mutants were obtained by sequencing analysis.Then,OsMT1 homozygous mutants were treated with NaCl solution with the concentrations of 50,100 and 150 mmol/L respectively.The results showed that the length of shoots and roots in OsMT1 mutant was significantly longer than that of wild type plants,indicating that OsMT1 mutants can decrease the salt sensitivity and promote the salt tolerance in rice.
