摘 要:食管癌是一种常见的消化道肿瘤,主要以手术和化疗为主,但复发率高.研究发现,青蒿素的衍生物蒿甲醚(artemether)能够抑制癌细胞增殖.本研究以食管癌细胞株ECA-109为研究对象,以二甲基亚砜(DMSO)为对照组,通过CCK-8方法检测蒿甲醚在不同作用浓度(0.1μM、1μM、10μM、100μM)以及不同时间梯度(2 h、4 h、6 h、24 h)对ECA-109细胞增殖的作用,使用酶标仪检测细胞OD值,通过GraphPad Prism软件进行数据分析.实验结果表明,蒿甲醚在100μM浓度时处理食管癌细胞株ECA-1092 h以及4 h后对癌细胞增殖有显著抑制作用(P<0.05);且药物处理4h后对癌细胞增殖抑制作用效果最好.本研究通过探索蒿甲醚对食管癌细胞株ECA-109增殖的抑制作用,为该疾病在临床上的治疗提供理论依据.Esophageal cancer is a common digestive tract tumor,and surgery and chemotherapy are the common treatment.However,the recurrence rate is high.Studies have found that ar⁃temether,a derivative of artemisinin,can inhibit the proliferation of cancer cells.In this study,esophageal cancer cells ECA-109 were used as the research object and dimethyl sulfoxide(DMSO)was used as the control group.The effects of artemether on the proliferation of ECA-109 cells at different concentrations(0.1μM,1μM,10μM,100μM)and different time gradi⁃ents(2 hours,4 hours,6 hours,8 hours)were detected by CCK-8 method.The OD value was detected by Microplate Reader,and the data were analyzed by GraphPad Prism software.The experimental results show that when the concentration of artemether is 100μM,and after treat⁃ment for 2hours and 4hours,it had a significant inhibitory effect on the proliferation of cancer cells(P<0.05),and artemether had the best inhibitory effect on the proliferation of esophageal cancer cells ECA-109 after treatment for 4hours.In this study,we explore the inhibitory effect of artemether on the proliferation of esophageal cancer cell line ECA-109,which provides a theoretical basis for the clinical treatment of this disease.
