摘 要:目的:参照STAIR技术标准,进行天麻提取物(GE2-1)促永久性脑缺血(pMCAO)模型大鼠神经功能俢复的研究。方法:线栓法复制大鼠pMCAO模型,分别于术后连续给予GE2-1至28 d,于造模后7,14,21,28 d,以行为学变化、脑组织病理改变(HE染色)和神经元损伤程度(Nissl染色)为主要观察指标,评价GE2-1改善缺血性脑卒中预后的作用,为其药效学成药性提供实验依据。结果:GE2-1干预14 d后能明显改善pMCAO模型大鼠的神经功能缺失症状,降低行为学评分,并能增加pMCAO模型大鼠脑组织的神经细胞尼氏体数量、减轻脑组织缺血病理损伤程度;在急性期给予GE2-1也未见其出血风险。结论:GE2-1可促进pMCAO模型大鼠恢复期神经功能的修复,具有改善缺血性脑卒中预后的作用。OBJECTIVE To study the nerve function recovery of rats with pMCAO induced by Gastrodia extract(GE2-1)according to the STAIR technical standard.METHODS The pMCAO model of rats was replicated by suture method,and GE2-1 was continuously given to rats after operation for 28 d.On the 7 th,14 th,21 st and 28 th days after modeling,behavioral changes,pathological changes of brain tissue(HE staining)and neuronal damage degree(Nissl staining)were used as the main observation indexes to evaluate the effect of GE2-1 on improving the prognosis of ischemic stroke and provide experimental basis for its pharmacodynamics.RESULTS GE2-1 could obviously improve the neurological deficits,decrease the behavioral score,increase the number of neuronal Nissl bodies in brain tissue of pMCAO model rats,and alleviate the degree of ischemic pathological injury of brain tissue after 14 days of intervention.There was no risk of bleeding when GE2-1 was given in acute stage.CONCLUSION GE2-1 can promote the recovery of neurological function in pMCAO model rats and improve the prognosis of ischemic stroke.
