摘 要:为了研究不同浓度碱性蛋白酶在犬骨骼标本制作过程中去除软组织的效果,本试验利用0.1%、0.3%、0.5%、0.7%和0.9%浓度的碱性蛋白酶分解液分别对5只3~5岁犬的头骨、四肢骨、胸骨和肋骨及脊柱骨进行软组织去除试验,观察不同时间、不同浓度的蛋白酶分解液对不同部位骨骼的软组织的分解效果。结果显示:头骨、四肢骨、胸骨和肋骨在0.3%浓度的碱性蛋白酶分解液中分别作用48、66 h和60 h,脊柱骨在0.5%浓度的碱性蛋白酶分解液中作用60 h,所有软组织完全分解且骨质没有明显破坏,长骨的骨骺完好,颅骨的骨缝没有明显的分离现象,肋骨没有断裂。结果表明,0.3%和0.5%浓度的碱性蛋白酶分解液在犬骨骼标本制作中去除软组织效果好,安全有效,可操作性强且成本低廉,能替代传统氢氧化钠溶液进行软组织去除。In order to determine the effect of different concentrations of alkaline protease on the soft tissues removal during canine bone specimens preparation,alkaline protease solutions at 0.1%,0.3%,0.5%,0.7%and 0.9%were used to treat the skull,limbs,sternum,ribs and spine from 5 dogs of 3-5 year-old,the effects of different processing times and concentrations of alkaline protease solutions on the soft tissues removal in different bones were observed.The results showed that soft tissues from the skull,limbs,sternum and ribs decomposed after treatment with 0.3%concentration of alkaline protease solution for 48,66 h and 60 h,respectively,while that from spinal bone decomposed after treatment with 0.5%concentration of alkaline protease solution for 60 h,without obvious bone damage,including intact long bone epiphyseal,and absence of separation on skull bone seam and rib fracture.Thus,0.3%and 0.5%alkaline protease solutions allow safe,effective,operable and low-cost removal of soft tissues in the preparation of canine bone specimens,and offers a valuable alternative to the traditional sodium hydroxide solution for removing soft tissues.
