高脂饮食诱导肥胖对大鼠心脏、肝脏线粒体呼吸功能的影响

摘  要:为了明确高脂饮食诱导肥胖对大鼠心脏、肝脏线粒体呼吸功能的影响,本试验采用液相氧电极检测大鼠心脏、肝脏线粒体磷氧比(ADP/O)和呼吸控制率(RCR)。结果显示,心脏ADP/O与RCR表现出相似的趋势,在第16周和第18周时,饮食诱导肥胖(DIO)组和饮食诱导肥胖抵抗(DR)组的ADP/O和RCR均显著低于正常对照(CON)组(P<0.05);肝脏RCR仅在第16周时出现了DIO组显著低于CON组(P<0.05);肝脏ADP/O在第10周时DIO组有高于CON组的趋势(P=0.057),在第18周时DIO组显著低于DR组(P<0.05),且有低于CON组的趋势(P=0.093)。结果表明,高脂饮食诱导肥胖可引起大鼠心脏线粒体呼吸功能受损。To investigate the effect of high-fat diet-induced obesity on mitochondrial respiratory function in rat heart and liver,this study utilized liquid-phase oxygen electrode to detect the phosphorus-to-oxygen ratio(ADP/O)and respiratory control rate(RCR)of rat heart and liver mitochondria.The results showed that cardiac ADP/O and RCR displayed similar trends,with the diet-induced obesity(DIO)group and diet-induced obesity resistance(DR)group being significantly lower than the control(CON)group at weeks 16 and 18(P<0.05);liver RCR was significantly lower in the DIO group than in the CON group only at week 16(P<0.05);liver ADP/O was higher in the DIO group than in the CON group at week 10(P=0.057)and significantly lower in the DIO group than in the DR group(P<0.05)and lower than in the CON group(P=0.093)at week 18.The results suggest that high-fat diet-induced obesity can impair mitochondrial respiratory function in rat hearts.
