摘 要:目的观察升清胶囊六种单体对H_(2)O_(2)诱导的人肝细胞LO2氧化应激的影响。方法将正常人肝细胞LO2分为正常组、模型组、白藜芦醇苷治疗组、白藜芦醇治疗组、橙皮苷治疗组、大黄酚治疗组、大黄素治疗组、大黄酸治疗组和维生素E对照组;模型组、治疗组、对照组建立H_(2)O_(2)造成人肝细胞LO2的氧化应激模型;治疗组及对照组每孔分别加入200μL含有不同浓度的白藜芦醇苷、白藜芦醇、橙皮苷、大黄酚、大黄素、大黄酸、维生素E的RPMI-1640培养液,正常组和模型组加入等体积RPMI-1640培养液,培养24 h。采用MTT法观察各组对H_(2)O_(2)造成人肝细胞氧化应激的影响,并分析其安全性。结果H_(2)O_(2)对肝细胞的半数抑制浓度为300μm;升清胶囊六种单体的IC_(50)均>150μm;在25~100μm范围内六种单体均有一定的修复作用;橙皮苷、白藜芦醇和大黄酚对H_(2)O_(2)造成的肝细胞氧化应激有较好的修复作用,最佳修复浓度分别为100μm、50μm、50μm,维生素E最佳修复浓度为50μm;橙皮苷修复作用优于VE(P<0.05),白藜芦醇修复作用和VE相当(P>0.05),大黄酚修复作用次于VE(P<0.05)。结论H_(2)O_(2)对肝细胞的IC_(50)为300μm,升清胶囊六种单体对肝细胞的安全性都比较高,对H_(2)O_(2)造成的肝细胞氧化应激均有一定的修复能力,橙皮苷、白藜芦醇和大黄酚有较好的修复作用,其修复作用分别优于、相当、次于VE。Objective To observe the effects of the six monomers of Shengqing Capsules on the oxidative stress of human hepatocytes LO2 induced by H_(2)O_(2).Methods The normal hepatocytes LO2 were divided into a normal group,a model group,a resveratrol glycoside treatment group,a resveratrol treatment group,a hesperidin treatment group,a chrysophanol treatment group,an emodin treatment group,a rhein treatment group and a vitamin E control group.An oxidative stress model of human hepatocytes LO2 induced by H_(2)O_(2) was established in the model group,the treatment group and the control group.The treatment group and the control group were added with 200μL of RPMI-1640 culture medium containing different concentrations of resveratrol glycoside,resveratrol,hemperidin,chrysophanol,emodin,rhein and vitamin E in each well,while the normal group and the model group were added with equal volume of RPMI-1640 culture medium,cultured for 24 h.The MTT assay was used to observe the effect of each group on the oxidative stress of human hepatocytes caused by H_(2)O_(2),and their safety was analyzed.Results The half inhibitory concentration of H_(2)O_(2) on hepatocytes was 300μm.The IC_(50) of the six monomers in Shengqing Capsules were all greater than 150μm.All the six monomers had a certain repair effect in the range of 25-100μm.Hesperidin,resveratrol and chrysophanol had a better repair effect on the H_(2)O_(2)-induced oxidative stress in hepatocytes,and their optimal repair concentration was 100μm,50μm and 50μm,respectively,while the best repair concentration of vitamin E was 50μm.The repair effect of hesperidin was better than that of VE(P<0.05),and the repair effect of resveratrol was similar to that of VE(P>0.05),while the repair effect of chrysophanol was inferior to that of VE(P<0.05).Conclusion The IC_(50) of H_(2)O_(2) on hepatocytes is 300μm.The six monomers of Shengqing Capsules are all relatively safe for hepatocytes,and have a certain repair effect on the oxidative stress of hepatocytes caused by H_(2)O_(2).Hesperi
