摘  要:为了探讨在常规精液冷冻保护剂中添加不同浓度的牛血清白蛋白(BSA)对FGF5基因编辑绵羊精液冷冻的效果。用人工采精法进行精液采集,在常规的精液冷冻保护剂中分别添加浓度为0.2%、0.5%、1%的牛血清白蛋白(BSA),以不添加BSA的常规精液冷冻保护剂作为对照,利用液氮熏蒸法对精液进行冷冻,对解冻后精子的活率、活力、顶体完整性和膜完整性进行评价,筛选BSA最佳保护浓度。浓度为0.5%的BSA在精液冷冻-解冻后,精子的活率、活力、顶体完整率各项指标能显著高于对照组(0%)和其他浓度(0.2%、1%)(P<0.05),精子的畸形率显著低于对照组和实验组(P<0.05)。浓度为0.5%的BSA在FGF5基因编辑绵羊精液冷冻中保护效果最佳,精子的各项指标得到一定的改善。本研究为FGF5基因编辑绵羊及相关品种公羊精子的冷冻保存提供了一种新的途径和有益的参考。This experiment was to investigate the effect of adding different concentrations of bovine serum albumin(BSA)to conventional semen cryoprotectants on the freezing of semen of FGF5 gene-edited sheep.[Methods]:Semen was collected by artificial semen collection,BSA with concentrations of 0.2%,0.5%,and1%were added to the conventional semen cryoprotectant respectively,the conventional semen without BSA as the control,and the semen was frozen by liquid nitrogen fumigation.After thawing,the viability,motility,acrosome integrity,and membrane integrity were detected,and the optimal concentration was screened.The results showed that after freezing thawing,the indexes of sperm viability,motility and acrosome integrity of0.5%BSA were significantly higher than those of the control group and other experimental groups(P<0.05),and the sperm abnormality rate was significantly lower than that of the control group and experimental groups(P<0.05).This experiment screened out that BSA with a concentration of 0.5%has the best protective effect in the freezing of semen of FGF5 gene-edited sheep,and various indicators of sperm have been improved to a certain extent.This study provides a new way and useful reference for the cryopreservation of FGF5 gene editing sheep and related breeds of ram sperm.
