摘 要:目的明确猪供肺冷缺血保存24 h过程中动脉血管随保存时间延长其收缩和舒张功能变化,通过肺动脉的微血管调节能力了解供肺情况,从血管功能层面分析供肺变化,为进一步供肺的保存和维护提供参考依据。方法按标准方法获取的猪供肺于4℃的perfadex-plus中保存,分别于1、6、12、24 h在显微镜下获取内径约1.0 mm的无分支肺动脉血管环,于器官浴槽中测定血管环的收缩和舒张功能,用高K+溶液和u-46619进行收缩刺激,待稳定后再以累计法加入不同浓度的p物质和罂粟碱进行舒张功能测定。结果u-46619诱发的肺动脉血管环的收缩力分别为:1 h(23.53±1.15)mN、6 h(23.97±0.98)mN、12 h(23.44±1.38)mN、24 h(23.63±0.99)mN,P=0.688,差异无统计学意义;以u-46619诱发的稳定收缩为基准,p物质所引起的最大舒张百分比分别为:1 h(96.74±0.89)、6 h(95.37±0.85)、12 h(91.86±1.20)、24 h(85.08±0.83),P<0.05;罂粟碱均能诱发完全舒张,P=0.309,差异无统计学意义。结论冷缺血保存24 h猪肺动脉血管的平滑肌收缩功能无明显变化;肺动脉血管内皮依赖的舒张功能逐渐降低,非内皮依赖的舒张功能无明显变化。Objective To clarify the changes in the constriction and relaxation function of arterial vessels in pigs during the 24-hours preservation of cold ischemia in the donor lung,to understand the donor lung condition through the mi⁃crovessel regulation ability of the pulmonary artery,and to analyze the changes in the donor lung from the level of vascu⁃lar function,so as to provide a reference for further preservation and maintenance of the donor lung.Methods The pig lung obtained by standard method was stored in perfadex-plus at 4℃,and the unbranded pulmonary artery vascular ring with an inner diameter of about 1.0 mm was obtained under the microscope at 1 h,6 h,12 h and 24 h,respective⁃ly,and the contraction and relaxation function of the vascular ring was measured in the organ bath,and the contractile stimulation was performed with high K+solution and u-46619,and then different concentrations of Substance p and Pa⁃paverine were added by the cumulative method to determine the relaxation function.Results The smooth muscle con⁃traction of pulmonary artery vessels induced by u-46619 was 1 hour(23.53±1.15)mN,6 hours(23.97±0.98)mN,12 hours(23.44±1.38)mN,24 hours(23.63±0.99)mN,P=0.688,and the maximum relaxation percentage of Substance p was 1 h(96.74±0.89)as the benchmark,6 h(95.37±0.85),12 h(91.86±1.20),24 h(85.08±0.83),P<0.05;Papaverine can induce complete diastolic,P=0.309,the difference was not statistically significant.Conclusion There was no significant change in the smooth muscle contractile function of pulmonary artery vessels in the preservation of cold ischemia for 24 hours,the diastolic function of pulmonary artery vascular endothelium dependence gradually decreased,and the non-endothelial-dependent diastolic function did not change significantly.
