摘  要:目的:探讨神经生长因子(NGF)对七氟醚诱导的小鼠基底前脑胆碱能系统退变及认知功能损害的治疗作用。方法:将C57BL/6小鼠分为对照组(control)和神经生长因子治疗组NGF,利用吸入七氟醚方法建立小鼠全身麻醉模型。NGF治疗组小鼠经鼻通路每天给予NGF 1 ng/g治疗,连续14 d,对照组小鼠经鼻通路给予同等剂量生理盐水。利用Morris水迷宫检测小鼠学习记忆功能,免疫荧光染色检测基底前脑胆碱乙酰转移酶(ChAT)阳性的胆碱能神经元,Western Blot检测海马ChAT蛋白表达。结果:与对照组相比,NGF组小鼠平台潜伏期显著缩短,平台穿越频次显著增加,原平台所在象限停留时间显著延长。NGF组小鼠基底前脑胆碱能神经元数目显著增加,海马ChAT表达显著增加。结论:NGF可减轻七氟醚诱导的基底前脑胆碱能系统退变及认知障碍。Objective:To explore the therapeutic effects of nerve growth factor(NGF)on sevoflurane-induced basal forebrain cholinergic system degeneration and cognitive dysfunction.Methods:C57 BL/6 mice were divided into control and experimental groups and were administered sevoflurane to establish the animal model of general anesthesia.Mice in the experimental group and the control group were given NGF(1 ng/g)and equal amount of saline daily via intranasal pathway for 14 days,respectively.Learning and memory function was assessed by Morris water maze.Choline acetyltransferase(ChAT)-positive cholinergic neurons in the basal forebrain were evaluated by immunofluorescence.Protein levels of ChAT in the hippocampus were determined by Western Blot.Results:Compared with controls,mice treated with NGF showed shorter escape latency,less platform crossing and shorter time spent in target quadrant.Further,mice treated with NGF exhibited increased number of cholinergic neurons in the basal forebrain,and increased expression of ChAT in the hippocampus.Conclusion:NGF could alleviate sevoflurane-induced basal forebrain cholinergic system degeneration and cognitive dysfunction.
