摘  要:目的:探讨低氧暴露对小鼠海马神经元发育相关蛋白表达的影响,为低氧导致认知功能障碍机制的研究提供新的靶分子。方法:低氧处理小鼠海马神经元来源的HT22细胞系24 h和48 h,收集细胞样本进行转录组测序筛查低氧易感分子,real time RT-PCR检测HT22细胞中低氧相关分子纤维连接蛋白1(Fn1)、细丝蛋白C(FLNC)和细胞周期蛋白F(CCNF)mRNA的表达水平,Western Blot检测HT22细胞中Fn1、FLNC和CCNF蛋白的表达水平。结果:转录组测序筛选出低氧相关分子,低氧暴露后HT22细胞中Fn1和FLNC mRNA和蛋白水平显著降低。结论:低氧暴露可导致HT22细胞中Fn1和FLNC表达发生改变,可能是低氧诱导神经元损伤的靶分子。Objective:To investigate the effect of hypoxic exposure on the expression of development-related moleculas in HT22 murine hippocampal neuronal cells,providing new targets for the study of the mechanism of hypoxia-induced cognitive dysfunction.Methods:HT22 cells derived from mouse hippocampal neurons were treated with hypoxia for 24 and 48 h,and cell samples were collected for transcriptome sequencing to screen for hypoxia-susceptible molecules.The mRNA expression levels of hypoxia-related molecules on fibronectin 1(Fn1),filamin C(FLNC)and cyclin F(CCNF)in HT22 cells were detected by real time RT-PCR,and the corresponding proteins expression in HT22 cells were verified by Western Blot.Results:Hypoxia-related molecules were screened by transcriptomics,and the mRNA and protein levels of Fn1 and FLNC in hippocampal neurons of mice were significantly decreased after hypoxia exposure.Conclusion:Hypoxia can lead to changes in the expression of Fn1 and FLNC in HT22 cells,which may be target molecules of hypoxia-induced neuronal damage.
