摘  要:目的:探讨硫酸锌(ZnSO_(4))促进脊髓损伤(SCI)小鼠的神经修复作用及分子机制。方法:成年雌性C57BL/6小鼠48只按照随机数表法分为假手术组(sham)、脊髓损伤组(SCI)和硫酸锌治疗组(ZnSO_(4)),采用Allen’s打击法制备小鼠SCI模型,治疗组小鼠进行腹腔注射硫酸锌。运用BMS评分评定小鼠的运动功能,术后5 d采用Western Blot检测小鼠脊髓损伤区锌转运蛋白体3(ZnT3)和哺乳动物雷帕霉素靶蛋白(mTOR)蛋白表达,免疫组织化学染色检测小鼠脊髓损伤区域ZnT3和mTOR蛋白表达与定位。结果:与sham组比较,SCI组及ZnSO_(4)组小鼠术后各时间点BMS评分降低,术后第7 d起,ZnSO_(4)组BMS评分高于SCI组(P<0.05);硫酸锌治疗5 d后,ZnSO_(4)组小鼠ZnT3表达高于SCI组,并且mTOR表达低于SCI组,差异具有统计学意义(P<0.01)。结论:硫酸锌可以通过调控小鼠脊髓损伤后ZnT3和mTOR的表达水平促进脊髓损伤的修复。Objective:To explore the neuroprotective effects of zinc sulfate(ZnSO_(4))on function recovery after spinal cord injury(SCI)in mice.Methods:Forty-eight adult female C57 BL/6 mice were randomly chosen and divided into three groups:sham group(sham),spinal cord injury group(SCI),and zinc sulfate intervention group(ZnSO_(4)).After established the Allen’s model of SCI,ZnSO_(4) group received zinc sulfate injection by peritoneal cavity.Function recovery was tested with BMS scores,and the expressions of zinc transporter 3(ZnT3)and mammalian target of rapamycin(mTOR)were dynamically detected by Western Blot and immunohistochemical staining in five days respectively after SCI.Results:Compared with sham group,the BMS scores of SCI group and ZnSO_(4) group significantly decreased,BMS scores of ZnSO_(4) group was higher than that of SCI group from the 7 th day after SCI(P<0.05).After 5 days of zinc sulfate treatment,the expression of ZnT3 in ZnSO_(4) group was higher than that of SCI group,and the expression of mTOR was lower than that of SCI group(P<0.01).Conclusion:Zinc sulfate can regulate the expression of ZnT3 and mTOR after spinal cord injury in mice,and promote the motor function in mice.
