七种农药诱发赤麂(Muntiacus muntjak)离体细胞染色体畸变、姐妹染色单体交换和细胞周期动力学变化的效应

摘  要:化学农药对人体健康的直接和潜在的威胁,特别是其致畸、致癌、致突变效应,已引起人们深切的关注,以不同的检测系统研究农药潜在的遗传学效应,并积累这方面的资料,是很有必要的。我们选择四种有机磷杀虫剂——甲胺磷、乙酰甲胺磷、甲基对硫磷和马拉硫磷,三种农用杀菌剂——敌菌丹、菌核净和敌枯双,在培养的赤麂(Muntiacus muntjak)成纤维细胞上进行诱发染色体畸变、姐妹染色单体交换和细胞周期延迟效应试验。Possible mutagenic activities of 4 organophosphorus insecticides(methyl-parathion,methamidophos,acephate,malathion)and 3 agricultural fungicides(captafol,dimethachlon,dikushuang)in the culture of Red Muntjac(Muntiacus muntjak)diploid cells were investigated using assays for chromosome aberration,sister-chromatid exchanges(SCEs)and cell-cycle delay.The experimental results revealed that 4 organophosphorus insecticides induced several types of chromosomal aberration,though the rates of chromosomal aberration did not show any remarkable increase;but 3 of the 4 organophosphorus insecticides induced significant increase of frequencies of S C E,the induction in decreasing order of SCE was acephate,methyl-parathion,me-thamidophos;the activities of captafol and dikushuang were the strongest in inducing SCE and chromosomal aberration among 7 pesticides,captafol was stronger than dikushuang.The number of SCE and rate of chromosomal aberration induced by captafol arc as high as 60.52±2.23/cell and 29.59%res-pcctively at the highest dose group,the sister chromatids per cell exchange to a great degree and many types of chromosomal aberration were induced.However the effect on cell cycle delay was seen clearly only at dikushuang group.It may be considered that captafol and dikushuang are very strong chemical mutagens,while methyl-parathion,acephate and methamidophos are weak chemical mutagens,Malathion and dimethachlon show no mutagenieity on Red Muntjac cells in vitro under our experimental conditions.
