摘 要:目的探讨急性主动脉夹层(AAD)-主动脉瓣二叶畸形(BAV)患者体内特异性的基因或蛋白质的表达情况。方法选取2021年2月至6月于安徽医科大学第一附属医院心脏大血管外科就诊并接受治疗的5例AAD-BAV患者和6例单纯性BAV患者,分别入选实验组及对照组。使用数据非依赖采集(DIA)蛋白组学技术分析了两组患者主动脉组织的差异蛋白表达情况,并通过生物信息学手段明确了差异蛋白的生物学功能及参与的异常信号通路。结果共鉴定出467个上调蛋白和481个下调蛋白。基于GO、KEGG数据库的差异表达蛋白富集分析揭示了差异蛋白主要参与了黏着斑、ECM受体相互作用、补体和凝血级联、剪接体、氧化磷酸化等相关信号通路。结论AAD-BAV患者主动脉组织内差异蛋白表达具有独特特征,其中许多差异蛋白是疾病发生和进展的关键蛋白,为未来疾病的早期诊断、精准治疗提供了参考依据。Objective To investigate the expression of genes or proteins specific to the patients with both acute aortic dissection(AAD)and bicuspid aortic valve(BAV).Methods Five BAV patients with AAD and six single BAV patients who were diagnosed and treated at the Department of Cardiocvascular Surgery,the First Affiliated Hospital of Anhui Medical University from February 2021 to Jane 2022 were recruited and enrolled into the experimental and control groups respectively.Data-independent acquisition(DIA)proteomics technology was used to analyse the differentially expressed proteins(DEPs)in the aortic tissues of the two groups and bioinformatics was used to identify the biological functions of the DEPs and the abnormal signaling pathways in which they were involved.Results A total of 467 upregulated proteins and 481 downregulated proteins were identified.GO and KEGG enrichment analysis revealed that DEPs were mainly involved in the signaling pathways related to focal adhesion,ECM-receptor interaction,complement and coagulation cascades,spliceosome,and oxidative phosphorylation,etc.Conclusion DEPs in the aortic tissues of AAD-BAV patients are of the characteristics,many of which are key proteins for the disease onset and progression,and these novel findings can provide a basis for early diagnosis and targeted treatment of the disease in the future.
