摘 要:目的探究可溶性Jagged1/Fc对鼠骨骺干细胞Notch信号通路的激活及生物学特性的影响.方法将2周龄内SP大鼠离体培养获取大鼠骨骺干细胞,设置不同浓度(0.5μg/mL、1μg/mL、2μg/mL)的Jagged1/Fc对第3代骨骺干细胞进行干预诱导,体外培养,在倒置显微镜下观察细胞生长情况,记录细胞贴壁情况及时间、细胞形态变化及增殖情况,采用CCK-8法检测细胞增殖活性,及使用Western印迹检测Jagged1/Fc对骨骺干细胞增殖分化影响.结果经可溶性Jagged1/Fc干预诱导后,与对照组比较,实验组与对照组细胞形态变化基本类似,但浓度为0.5μg/mL的Jagged1/Fc实验组细胞增殖速度显著加快,贴壁所需的时间较对照组缩短(P<0.05).CollagenⅡ蛋白表达明显增多,胶原蛋白X表达明显减少.结论单个Jagged1/Fc配体可激活Notch信号通路,促进骨骺干细胞增殖的最适浓度是0.5μg/mL.Objective To investigate the effect of soluble Jagged1/Fc on the activation and biological characteristics of Notch signaling pathway in rat epiphyseal stem cells.Methods SP rats within 2 weeks-old were cultured in vitro to obtain rat epiphyseal stem cells.Jagged1/Fc with different concentrations(0.5 μg/mL, 1 μg/mL and 2 μg/mL) was set to intervene and induce the third generation epiphyseal stem cells.They were cultured in vitro.The cell growth, cell adhesion and time, cell morphological changes and proliferation were observed under inverted microscope, the cell proliferation activity was detected by CCK-8 method, and the effect of Jagged1/Fc on the proliferation and differentiation of epiphyseal stem cells was detected by Western blot.Results After the intervention and induction of soluble Jagged1/Fc, the morphological changes of cells in the experimental group were basically similar to those in the control group, but the proliferation rate of cells in the experimental group with jagged1/Fc at the concentration of 0.5 μg/mL was significantly faster and the time required for adhesion was shorter than that in the control group(P<0.05) The expression of collagen Ⅱ protein increased significantly and the expression of collagen X decreased significantly.Conclusion Single Jagged1/Fc ligand can activate Notch signaling pathway, and the optimal concentration to promote the proliferation of epiphyseal stem cells is 0.5 μg/mL.
