摘  要:目的探讨七氟烷预处理对糖尿病大鼠视网膜保护作用及其分子机制。方法将30只成年健康SD大鼠按照随机数字表随机分为对照组、糖尿病组和七氟烷组。糖尿病组和七氟烷组分别通过腹腔注射链脲佐菌素(STZ)建立糖尿病模型。七氟烷组大鼠给予连续4 d七氟烷处理。采用生化检测分析3组血糖变化;采用苏木精-伊红(HE)染色分析3组大鼠视网膜病理变化;采用比色法分析视网膜组织氧化应激指标变化;采用蛋白质免疫印迹分析3组大鼠视网膜组织内质网应激蛋白表达水平;采用原位缺口末端标记法(TUNEL)染色分析视网膜组织细胞凋亡水平;组间比较采用单因素方差分析。结果七氟烷组大鼠视网膜组织学评分[(1.53±0.22)分]明显低于糖尿病组[(3.28±0.16)分], 差异有统计学意义(t=20.111, P<0.05)。七氟烷组大鼠视网膜组织丙二醛(MDA)水平[(3.29±0.08) mmol/L]明显低于糖尿病组[(3.83±0.23) mmol/L], 差异有统计学意义(t=6.780, P<0.05)。七氟烷组大鼠视网膜组织超氧化物歧化酶(SOD)水平[(7.59±0.71) U/mg]明显低于糖尿病组[(9.74±0.58) U/mg], 差异有统计学意义(t=13.240, P<0.05)。七氟烷组大鼠视网膜组织SOD水平[(155.90±17.14)%]明显低于糖尿病组[(255.80±23.76)%], 差异有统计学意义(t=10.780, P<0.05)。七氟烷组大鼠视网膜组织葡萄糖调节蛋白78(GRP78)表达水平(1.46±0.12)明显低于糖尿病组(2.19±0.10), 差异有统计学意义(t=14.880, P<0.05)。七氟烷组大鼠视网膜组织CHOP蛋白表达水平(2.55±0.17)明显低于糖尿病组(2.16±0.29), 差异有统计学意义(t=3.720, P<0.05)。七氟烷组大鼠视网膜组织凋亡比例[(15.01±1.54)%]明显低于糖尿病组[(23.10±3.17)%], 差异有统计学意义(t=7.260, P<0.05)。结论七氟烷处理可显著下调糖尿病大鼠视网膜细胞的氧化应激和内质网应激, 降低视网膜细胞凋亡, 进而对糖尿病大鼠视网膜起保护作用。Objective To investigate the protective effect of sevoflurane pretreatment on the retina of diabetes rats and its molecular mechanism.Methods Totally,30 healthy adult SD rats were randomly divided into control group,diabetes group and sevoflurane group.Diabetes group and sevoflurane group were established by intraperitoneal injection of streptozotocin(STZ).Rats in sevoflurane group were treated with sevoflurane for 4 days.The changes of blood glucose in the three groups were analyzed by biochemical detection.The pathological changes of retina in the three groups were analyzed by hematoxylin and eosin(HE)staining.The changes of oxidative stress in retina were analyzed by colorimetry.The expression of ER stress protein in retina of three groups was analyzed by Western blotting.The level of retinal cell apoptosis was analyzed by terminal-deoxynucleotidyl transferase mediated nick end labeling(TUNEL)staining.One way ANOVA was used for comparison between groups.Results The histological score of retina in sevoflurane group[(1.53±0.22)points]was significantly lower than that in diabetes group[(3.28±0.16)points,t=20.111,P<0.05].The malondialdehyde(MDA)level of retina in sevoflurane group[(3.29±0.08)mmol/L]was significantly lower than that in diabetes group[(3.83±0.23)mmol/L,t=6.780,P<0.05].The level of superoxide dismutase(SOD)in retina of rats in sevoflurane group[(7.59±0.71)U/mg]was significantly lower than that in diabetes group[(9.74±0.58)U/mg,t=13.240,P<0.05].The SOD level of retina in sevoflurane group[(155.90±17.14)%]was significantly lower than that in diabetes group[(255.80±23.76)%,t=10.780,P<0.05].The expression level of glucose regulated P rotein 78 kD(GRP78)in retina of rats in sevoflurane group(1.46±0.12)was significantly lower than that in diabetes group(2.19±0.10,t=14.880,P<0.05).The expression level of chop protein in retina of rats in sevoflurane group(2.55±0.17)was significantly lower than that in diabetes group(2.16±0.29,t=3.720,P<0.05).The apoptosis rate of retinal tissue in sevoflurane gr
