电针对营养性肥胖雌性生长期大鼠糖脂代谢相关指标及脂肪组织形态的影响

摘　　要：目的:观察电针对高脂饮食诱导的雌性生长期大鼠糖脂代谢功能的影响。方法:21 d龄SD雌性大鼠随机分为对照组6只给予普通饲料喂养,模型组6只给予高脂饲料喂养且不做任何治疗,电针组6只高脂饲料喂养并选取“三阴交”“丰隆”“足三里”给予电针治疗,连续干预14 d。每周测量大鼠体质量和腹围,治疗结束后分别采用全自动生化检测和ELISA检测血清总胆固醇(total cholesterol,TC)、甘油三酯(triglyceride,TG)、高密度脂蛋白胆固醇(high density lipoprotein cholesterol,HDL-C)、低密度脂蛋白胆固醇(low-density lipoprotein cholesterol,LDL-C)、血糖(blood glucose,GLU)和血清胰岛素(insulin,INS)、瘦素(leptin,LEP)水平,采用HE染色法观察肾周脂肪组织形态。结果:与对照组比较,模型组体质量明显增加(P<0.05),血清TC、TG含量明显升高(P<0.05),血清HDL-C含量明显降低(P<0.05),血清INS、GLU、LEP含量明显升高(均P<0.05),肾周脂肪细胞形态饱满、体积增大,细胞内可见空泡状脂滴;与模型组比较,电针组体质量明显降低、腹围减小(P<0.05),血清TC、TG明显降低(P<0.05),血清INS、GLU、LEP含量明显降低(均P<0.05),肾周脂肪细胞体积缩小,胞体皱缩。结论:电针可以改善高脂饮食诱导的生长期雌性肥胖大鼠的糖脂代谢功能,降低胰岛素和瘦素水平,改善肾周脂肪的组织形态。Objective:To observe the effects of electroacupuncture on glucose and lipid metabolismin female growing rats induced by high-fat diet.Methods:21-day-old female SD rats,6 rats were selected as the control group and fed with ordinary feed,6 rats were selected as model group and fed with high-fat diet without any treatment,6 rats were selected as the electroacupuncture group and fed with high-fat diet,as well as treated with electroacupuncture on Sanyinjiao(SP6),Fenglong(ST40)and Zusanli(ST36).During the intervention period,the body mass and abdominal circumference of rats were measured every week.After 2 weeks of treatment,serum cholesterol(TC),triglyceride(TG),high density lipoprotein(HDL-C),low-density lipoprotein(LDL-C)and blood glucose(GLU)were detected by automatic biochemical detection,serum insulin(INS)and leptin(LEP)were detected by ELISA.The morphology of perirenal adipose tissue were observed by HE staining.Results:Compared with the control group,the body mass of the model group increased significantly(P<0.05),the levels of serum TC and TG increased significantly(P<0.05),serum HDL-C decreased significantly(P<0.05),the levels of serum INS,GLU and LEP increased significantly(P<0.05),the adipocytes around the kidney were plump and large,and vacuolar lipid droplets could be seen in the cells.Compared with the model group,the body mass(P<0.05),abdominal circumference(P<0.05),serum TC and TG levels decreased significantly(P<0.05),serum INS,GLU and LEP levels decreased significantly(P<0.05),the cell volume of perirenal adipose tissue decreased and the cell body shrank.Conclusion:Electroacupuncture can improve the glucose and lipid metabolism of growing female obese rats induced by high-fat diet,reduce insulin,leptin,and improve the tissue morphology of perirenal fat around kidney.
