摘　　要：目的观察COX-2抑制剂对于尿道损伤中TGF-β1表达的影响。方法雄性SD大鼠24只随机分为3组,每组8只。A组为空白对照组,B组为模型组,C组为实验组。手术制备尿道狭窄模型后,分别定时定量灌注生理盐水、COX-2抑制剂共6个月。6个月后处死实验鼠,采集尿道组织进行病理组织学检查。结果使用Masson染色检测胶原纤维,3组数据比较,差异有统计学意义(P<0.01)。胶原纤维含量B组>C组>A组。ELISA检测3组间样本TGF-β1实际浓度的差异性比较,差异有统计学意义(P<0.01)。Western blot检测TGF-β1蛋白表达结果显示,3组TGF-β1蛋白表达差异有统计学意义,B组>C组>A组。结论实验鼠术后给予COX-2抑制剂治疗,可下调TGF-β1的表达,从而减少尿道狭窄的发生或延长尿道狭窄发生的时间。Objective To observe the effect of COX-2 inhibitor on the expression of TGF-β1 in urethral injury.Methods Twenty-four male SD rats were randomly divided into 3 groups,8 in each group.Group A was a blank control group,group B was a model group,and group C was an experimental group.After surgically preparing the urethral stricture model,they were infused with normal saline and COX-2 inhibitor regularly and quantitatively for 6 months.After 6 months,the experimental mice were sacrificed,and urethral tissue was collected for histopathological examination.Results Masson staining was used to detect collagen fibers,which showed that the P value among the three groups was statistically significant(P<0.01).Collagen fiber content were group B>group C>group A.ELISA detected the difference in the actual concentration of the sample TGF-β1 among the three groups.The results showed that the P value was less than 0.01,which was statistically significant.Western blot detection of TGF-β1 protein expression results showed that the three groups of TGF-β1 protein expression were different:group B>group C>group A.Conclusion Postoperative treatment of experimental mice with COX-2 inhibitor can down-regulate the expression of TGF-β1,thereby reducing the occurrence of urethral stricture or prolonging the occurrence time of urethral stricture.
