摘 要:目的 观察肌腱蛋白(TN)-X、TN-C及转化生长因子β(TGF-β)在心肌纤维化过程中的表达变化及其与纤维化严重程度的相关性。方法 选择雄性SD大鼠48只,随机分为模型组43只和对照组5只。模型组建立心肌纤维化模型,采用Masson染色观察心肌组织基本结构,计算胶原容积分数(CVF),采用免疫组织化学法检测心肌中TN-X、TN-C及TGF-β表达。结果 与对照组比较,模型组建模后2、5、8、11、14 d CVF明显升高[(19.13±2.83)%、(21.43±1.45)%、(26.68±2.36)%、(30.69±1.01)%、(30.87±0.73)%vs(2.36±0.29)%,P<0.01]。模型组建模后5、8、11 d CVF呈上升趋势(P<0.05)。免疫组织化学显示,模型组建模后5、8、11、14 d TGF-β和TN-X及2、5、8 d TN-C表达明显高于对照组(P<0.05,P<0.01)。TGF-β、TN-X与CVF呈正相关(r=0.701,r=0.752,P<0.01);TN-X与TGF-β呈正相关(r=0.764,P<0.01)。结论 TN-X和TGF-β随着心肌纤维化严重程度增加而增加;TN-X、TN-C可能在心肌纤维化的不同阶段起调控作用。Objective To investigate the expression profiles of TN-X,TN-C and TGF-β in the formation of myocardial fibrosis and their relationship with the formation.Methods Forty-eight male SD rats were randomly divided into model group(n=43) and normal control group(n=5).Rat model of myocardial fibrosis was induced in model group.Masson staining was used to observe the basic structure of the myocardium, and histological collagen volume fraction(CVF) was then calculated.The expression of TN-X,TN-C and TGF-β was determined by immunohistochemical assay.Results The CVF of the model group was significantly higher than that of the control group at 2,5,8,11 and 14 d after modeling [(19.13±2.83)%,(21.43±1.45)%,(26.68±2.36)%,(30.69±1.01)% and(30.87±0.73)% vs(2.36±0.29)%,P<0.01].The CVF was in a trend of rising at 5,8 and 11 d after modeling(P<0.05).The results of immunohistochemical assay showed that the expression levels of TGF-β and TN-X at 5,8,11 and 14 d after modeling, and that of TN-C at 2,5 and 8 d after modeling were remarkably higher than those of the control group(P<0.05,P<0.01).The expression levels of TGF-β and TN-X were significantly correlated with CVF(r=0.701,r=0.752,P<0.01);and the expression of TN-X had a positive correlation with that of TGF-β(r=0.764,P<0.01).Conclusion With the severity of myocardial fibrosis, the expression of TN-X and TGF-β is increased gradually.TN-X and TN-C may play regulative roles in the different stages of myocardial fibrosis.
