摘 要:目的 探讨月见草油对蛛网膜下腔出血(SAH)大鼠表皮生长因子受体(EGFR)/信号转导和转录激活因子3(STAT3)通路及运动功能的影响。方法 将60只大鼠分为假手术组(假手术+灌胃生理盐水)、SAH组(SAH建模+灌胃生理盐水)及月见草油低、中、高剂量组(SAH建模+灌胃135 mg/kg、270 mg/kg、540 mg/kg月见草油)。对大鼠进行运动神经功能评分,苏木精-伊红(HE)染色、末端脱氧核苷酸转移酶介导的dUTP缺口末端标记(TUNEL)染色观察脑组织病理损伤及脑组织细胞凋亡,酶联免疫吸附实验(ELISA)检测脑组织中白细胞介素(IL)-1β、IL-6、肿瘤坏死因子-α(TNF-α)表达水平,蛋白质印迹法(Western Blot)检测脑组织中EGFR、磷酸化STAT3(p-STAT3)、STAT3蛋白表达水平。结果 与假手术组相比,SAH组大鼠运动神经功能评分明显降低(P<0.05),脑组织细胞凋亡率、脑组织中IL-1β、IL-6、TNF-α水平及EGFR、p-STAT3/STAT3蛋白表达水平明显升高(P<0.05),且大脑皮层神经细胞有严重坏死迹象。随着月见草油的治疗干预及干预剂量的升高,SAH大鼠运动神经功能评分逐渐升高(P<0.05),脑组织细胞凋亡率、脑组织中IL-1β、IL-6、TNF-α水平及EGFR、p-STAT3/STAT3蛋白表达水平逐渐降低(P<0.05),且大脑皮层神经细胞的形态结构损伤有不同程度的减轻。结论 月见草油可抑制SAH大鼠脑组织中EGFR/STAT3通路激活,降低大鼠脑组织中炎症水平及细胞凋亡率,促进运动功能恢复。Objective To investigate the effects of Evening Primrose Oil on epidermal growth factor receptor(EGFR)/signal transducer and activator of transcription 3(STAT3) pathway and motor function in subarachnoid hemorrhage(SAH) rats.Methods The rats were divided into sham operation group(sham operation+intragastric administration of normal saline),SAH group(SAH modeling+intragastric administration of normal saline),and low-,medium-,and high-dose Evening Primrose Oil groups(SAH modeling+intragastric administration of 135 mg/kg, 270 mg/kg, 540 mg/kg Evening Primrose Oil, respectively).The motor function of rats was evaluated.Hematoxylin and eosin staining and Terminal-deoxynucleoitidyl Transferase Mediated Nick End Labeling(TUNEL) staining were used to observe the pathological damage and apoptosis of brain tissue.Enzyme-linked immunosorbent assay(ELISA) was used to detect the expression levels of interleukin(IL)-1β,IL-6,and tumor necrosis factor-α(TNF-α).Western Blot was used to detect the protein expression of EGFR,phosphorylated STAT3(p-STAT3),and STAT3 in brain tissue.Results Compared with those in sham operation group, the motor nerve function score in SAH group was significantly decreased(P<0.05),the apoptosis rate in brain tissue, levels of IL-1β,IL-6,TNF-α,protein levels of EGFR and p-STAT3/STAT3 in brain tissue were significantly increased(P<0.05),there were signs of severe necrosis in the cerebral cortex.With the therapeutic intervention and increase of intervention dose of Evening Primrose Oil, the motor nerve function scores of SAH rats were increased gradually(P<0.05),the apoptosis rate in brain tissue, levels of IL-1β,IL-6,TNF-α,protein levels of EGFR and p-STAT3/STAT3 in brain tissue were decreased gradually(P<0.05),the damage of structure of the neurons in the cerebral cortex was alleviated in varying degrees.Conclusion Evening Primrose Oil can inhibit the activation of EGFR/STAT3 pathway in brain tissue of SAH rats, reduce the level of inflammation and the rate of apoptosis, and promote the recovery
