摘  要:目的探讨丹红注射液对老龄大鼠麻醉术后早期认知功能障碍的影响。方法选取30只SPF级、健康、老龄、雄性SD大鼠,采用随机区组法随机分为对照组、术后认知功能障碍(post operative cognitive dysfunction,POCD)组和实验组,每组各10只。POCD组和实验组大鼠通过探查腹腔脏器的方法建立POCD模型,实验组大鼠于手术结束前30min及术后第1、2天分别予2ml/kg丹红注射液(Danhong injection,DHI),POCD组大鼠在相同时间点予相同剂量的生理盐水,对照组不进行手术,但在相同时间点予相同剂量的生理盐水。采用Morris水迷宫实验评价大鼠的认知功能;采用酶联免疫吸附试验(enzyme-linked immunoadsordent assay,ELISA)检测血清S-100β蛋白、神经元特异性烯醇化酶(neuron specific enolase,NSE)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-6(interleukin-6,IL-6)和白细胞介素-1β(interleukin-1β,IL-1β)水平。结果DHI能够缩短POCD大鼠的逃逸潜伏期,增加穿越平台次数,提高POCD大鼠的学习记忆能力;同时不同实验组S-100β蛋白水平[术后6h(1.05±0.03)、术后24h(0.98±0.15)、术后48h(0.79±0.05)]较POCD组[(术后6h(1.39±0.22)、术后24h(1.24±0.13)、术后48h(1.19±0.15)]降低,实验组NSE水平[术后6h(15.71±5.12)、术后24h(13.35±4.64)、术后48h(12.59±4.22)]低于POCD组[术后6h(25.49±4.82)、术后24h(20.71±5.12)、术后48h(18.43±3.72)];POCD组中TNF-α(112.6±16.7)、IL-6(39.6±6.5)、IL-1β(39.5±6.7)水平均较实验组TNF-α(71.2±15.3)、IL-6(30.4±5.6)、IL-1β(23.0±5.6)高。结论DHI能够提高POCD大鼠的学习记忆能力,减轻POCD大鼠的神经系统损伤及POCD导致的炎性反应,改善老龄大鼠麻醉术后早期认知功能障碍。Objective To explore the effect of Danhong injection(DHI)on early cognitive dysfunction in aged rats after anesthesia.Methods Thirty SPF grade,healthy,aged,male SD rats were randomly divided into control group postoperative cognitive dysfunction(POCD)group and experimental group used random block method,with 10 rats in each group.The rats of POCD group and experiment group established postoperative cognitive dysfunction(POCD)model by probing abdominal organs.The rats of experimental group was given DHI at a dose of 2ml/kg on 30min before the end of operation,and the 1st and 2nd day after operation,the rats of POCD group received the same dose of normal saline at the same time point,while the control group was not undergo surgery but given the same dose of saline at the same time point.The cognitive function of rats was evaluated by Morris water maze test.The contents of S-100βprotein,neuron specific enolase(NSE),tumor necrosis factorα(TNF-α),interleukin-6(IL-6)and interleukin-1β(IL-1β)were determined by enzyme-linked immunoadsordent assay(ELISA).Results DHI can shorten the escape latency of POCD rats,increase the frequency of crossing the platform,improve the learning and memory ability of POCD rats.The levels of S-100βprotein in the experimental group[(1.05±0.03)at 6h,(0.98±0.15)at 24h,(0.79±0.05)at 48h after operation]were lower than those of POCD[(1.39±0.22)at 6h,(1.24±0.13)at 24h,1.19±0.15 at 48h after operation]group.NSE level also obtained similar results.The experimental group[(15.71±5.12)at 6h,(13.35±4.64)at 24h,(12.59±4.22)at 48h after operation]was lower than the POCD group[(25.49±4.82)at 6h,(20.71±5.12)at 24h,(18.43±3.72)at 48h after operation].The levels of TNF-α(112.6±16.7),IL-6(39.6±6.5)and IL-1β(39.5±6.7)in POCD group were higher than those in experimental group[TNF-α(71.2±15.3),IL-6(30.4±5.6)and IL-1β(23.0±5.6)].Conclusion DHI can improve the learning and memory ability of POCD rats,reduce the damage of the nervous system of POCD rats and the inflammatory reaction cause
