摘 要:为探究铬(Cr)胁迫下添加外源褪黑素(MT)对百日草(Zinnia elegans)种子萌发及幼苗生理的缓解效应,采用不同浓度(0、50、100、200、400、600μmol/L)MT处理100μmol/L Cr胁迫下的百日草种子和幼苗,统计种子萌发状况,并测定幼苗相关生理生化指标。结果表明,Cr浓度为100μmol/L时对百日草种子萌发和幼苗生长造成严重伤害;施加外源MT可以提高Cr胁迫下百日草种子的发芽指标,增加幼苗可溶性糖、可溶性蛋白和叶绿素含量,降低丙二醛(MDA)含量,增强超氧化物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶(CAT)活性。当MT浓度为100μmol/L时,缓解Cr胁迫效果最佳,与单一Cr胁迫处理相比,百日草种子发芽率、发芽势、发芽指数和活力指数分别升高27.42%、37.35%、46.39%和82.63%,幼苗可溶性糖、可溶性蛋白、叶绿素a、叶绿素b和总叶绿素含量分别增加49.53%、54.14%、62.91%、136.50%和79.71%,MDA含量降低49.49%,SOD、POD和CAT活性分别提高43.89%、119.95%和65.49%。可见,Cr胁迫下增施外源MT可促进百日草种子萌发,缓解Cr胁迫对幼苗细胞膜系统的损伤,提高渗透调节能力,增强抗氧化酶活性,从而提高百日草对Cr胁迫的抗性。The effects of exogenous melatonin(MT)on seed germination and seedling physiological characters of Zinnia elegans under chromium(Cr)stress were studied to provide theoretical bases for relieving the toxic of Cr stress.Different concentrations of MT(0,50,100,200,400,600μmol/L)were used to treat seeds and seedling of Z.elegans under 100μmol/L Cr stress.The seed germination status was investigated and the related physiological and biochemical indexes of seedlings were measured.The results showed that seed germination and seedling growth of Z.elegans were damaged significantly under 100μmol/L Cr stress.Exoge⁃nous MT could improve the germination index,increase the contents of soluble sugar,soluble protein and chlorophyll,reduce the MDA content,and enhance the activities of superoxide dismutase(SOD),peroxidase(POD)and catalase(CAT).The relieving effect was the best when MT was 100μmol/L,and compared with single Cr treatment,the germination percentage,germination potential,germination index and vital index were improved by 27.42%,37.35%,46.39%and 82.63%,respectively;the contents of soluble sugar,soluble protein,chlorophyll a,chlorophyll b and total chlorophyll were increased by 49.53%,54.14%,62.91%,136.50%and 79.71%,respectively;the contents of MDA were reduced by 49.49%;the activities of SOD,POD and CAT were enhanced by 43.89%,119.95%and 65.49%,respectively.It was concluded that exoge⁃nous MT application promoted the seed germination of Z.elegans under Cr stress,and alleviated the damage to seedling cell membrane system,improved the osmotic adjustment ability,and enhanced the antioxidant en⁃zyme activities,thereby improved the stress resistance to Cr stress.
