摘 要:为了解山东地区蛋鸡源大肠杆菌中bla_(CTX-M)型超广谱β-内酰胺酶耐药基因的流行特征,本研究分析了2019年从山东不同地区共5个蛋鸡养殖场分离的440株大肠杆菌中bla_(CTX-M)基因的流行情况,并分析其耐药性及可转移性。结果表明:142株(32.3%)大肠杆菌携带bla_(CTX-M)基因,包括bla_(CTX-M)-1G(20.2%)、bla_(CTX-M-9G)(14.1%)。药敏试验结果显示,携带bla_(CTX-M)基因的大肠杆菌对阿莫西林的耐药率最高(95.8%),对氨苄西林耐药率次之(94.4%),对萘啶酸、头孢噻肟、头孢噻呋、恩诺沙星、氟苯尼考耐药水平较高,介于82.4%~86.6%之间,对阿米卡星的耐药率最低(2.8%)。对携带bla_(CTX-M)基因的大肠杆菌进行接合转移试验,结果显示,142株bla_(CTX-M)阳性菌株中有139株接合转移成功,复制子分型主要为F型,占82.0%。本研究结果说明山东地区蛋鸡源大肠杆菌耐药情况较为严重,bla_(CTX-M)基因主要以F型质粒作为接合转移载体进行水平传播,加重了耐药基因的传播及多药耐药情况。To understand the prevalence of bla_(CTX-M)extended⁃spectrumβ⁃lactamase resistance genes in Escherichia coli from laying hens in Shandong region,we analyzed the prevalence of bla_(CTX-M)gene of 440 E.coli strains isolated from five laying hen farms in different regions of Shandong in 2019,and analyzed its drug resist⁃ance and transferability.The results showed that 142 strains(32.3%)of E.coli carried the bla_(CTX-M)gene,inclu⁃ding bla_(CTX-M)⁃1G(20.2%)and bla_(CTX-M⁃9G)(14.1%).The drug susceptibility test results showed that the E.coli car⁃rying the bla_(CTX-M)gene had the highest resistance rate to amoxicillin(95.8%),followed by ampicillin(94.4%);the resistance rate to nalidixic acid,cefotaxime,ceftiofur,enrofloxacin and florfenicol were higher ranging from 82.4%to 86.6%,and that to amikacin was the lowest(2.8%).Conjugation transfer test was performed on the E.coli carrying bla_(CTX-M)gene.The results showed that 139 of the 142 bla_(CTX-M)positive strains were successfully conjugated and transferred,and the replicon type was mainly F,accounting for 82.0%.The results of this study indicated that the drug resistance of E.coli from laying hens in Shandong was relatively serious.The bla_(CTX-M)gene mainly used F plasmid as a conjugative transfer vector for horizontal transmission,which aggravated the spread of drug resistance genes and multidrug resistance.
