摘 要:目的:建立沉香挥发油的薄层色谱鉴别方法,为其品质评价提供依据。方法:以苄基丙酮为对照品,对薄层色谱展开系统、展开过程、展距进行优化和方法学验证。结果:自制硅胶G薄层板,常温下二次展开,展开剂Ⅰ:正己烷-丙酮(95︰5)上行展开8cm,展开剂Ⅱ:石油醚(60~90℃)-乙酸乙酯(95︰5)上行展开18 cm,5%香草醛硫酸乙醇溶液为显色剂,105℃加热显色后日光下检视,薄层色谱斑点清晰、特征性好。结论:该方法操作简便、直观性强,专属性、稳定性和耐用性好,可用于水蒸气蒸馏制取的沉香挥发油鉴别。Objective:To establish the thin-layer chromatography(TLC)identification method for agarwood oil and provide the basis for the quality evaluation.Methods:Using benzylacetone as a reference substance,the development system,development process,and development distance of TLC were optimized.The methodological investigation of the determined TLC conditions was carried out.Results:With the method of secondary separation of the TLC,the sample solution was developed on the self-made 0.4 mm of silica G thin layer plate at room temperature,when n-hexane-acetone(95︰5)was used as the first developing solvent and the petroleum ether(60~90℃)-ethyl acetate(95︰5)was used as the second developing solvent,and then sprayed with 5%vanillin-sulfuric acid ethanol solution and heated to color in 105℃for the view under daylight.The obtained separation effect of TLC for agarwood oil was good,the spots were clear,and the characteristics were significant.Conclusion:The method is easy to operate,highly intuitive,and has good specificity,stability and durability,and can be used for the identification of agarwood volatile oil prepared by steam distillation.
