摘 要:目的探究右美托咪啶对大鼠缺血/再灌注损伤肺组织核因子-κB(NF-κB)表达的调控及对缺血/再灌注损伤肺的保护机制。方法随机将SD大鼠分为假手术组(sham组)、缺血/再灌注组(I/R组)、右美托咪啶组(Dex组)、阿替美唑组(Atip组)与右美托咪啶+阿替美唑组(Dex+Atip组)。在给药结束后,处死大鼠取出左肺组织,检测总肺水含量(TLW)和肺湿干重比(W/D);电镜观察大鼠肺组织超微结构;RT-PCR检测肺组织NF-κB mRNA表达;WB检测肺组织NF-κB的蛋白表达。结果对比于I/R组,Dex组电镜下肺组织损伤轻,NF-κB mRNA、蛋白表达量、TLW与W/D均减少(P<0.05);而I/R组、Atip组和Dex+Atip组3组的差异无统计学意义。结论右美托咪啶可能通过抑制NF-κB表达来缓解肺缺血/再灌注损伤。OBJECTIVE To investigate the effect of Dexmedetomidine on NF-κB expression in lung during lung ischemia/reperfusion in rats and its possible protecting mechanisms.METHODS Sprague-Dawley rats were randomly divided into five groups:sham-operated group(sham group),ischemia/reperfusion group(I/R group),dexmedetomidine group(Dex group),atipamezole group(Atip group),dexmedetomidine+atipamezole group(Dex+Atip group).After the experiment the rats were killed and the left lung tissues were harvested to determine the total lung water content(TLW)and lung wet/dry weight(W/D);Ultra structure of lung tissues were observed under electron microscopy.The mRNA expression of nuclear factor-kappa B(NF-κB)in lung tissues were determined by RT-PCR;The protein level of NF-κB in lung tissues were detected by western blot.RESULTS Compared with I/R group,the structure damages of lung tissues observed under electron microscopy in Dex group were slighter,the mRNA and protein expression of NF-κB in lung tissues were decreased(P<0.05),W/D and TLW in Dex group was lower(P<0.05).There was no significant difference of the above parameters among I/R group,Atip group,and Dex+Atip group.CONCLUSION Dexmedetomidine can alleviate the lung ischemia/reperfusion injury through inhibiting NF-κB expression.
