摘 要:目的考察刺苋根不同部位提取物在体外对草酸钙致人肾小管上皮细胞(HK-2)损伤的保护作用。方法以氯化钙和草酸钠损伤HK-2细胞模型体外筛选,以CCK-8法检测刺苋根不同部位提取物对草酸钙致HK-2细胞损伤的保护作用,并通过HE染色,观察HK-2细胞形态。结果刺苋根不同部位提取物可不同程度提高草酸钙损伤后细胞的存活率,2、1、0.5、0.25 mg·mL^(-1)醇提取液组细胞存活率分别为(105.80±0.08)%、(111.79±0.01)%、(121.43±0.20)%、(128.92±0.00)%;0.5、0.25 mg·mL^(-1)石油醚提取液组细胞存活率分别为(112.72±0.45)%、(99.94±0.28)%;1、0.5、0.25 mg·mL^(-1)乙酸乙酯提取液组细胞存活率分别为(14.04±0.03)%、(111.24±0.08)%、(121.37±0.03)%;1、0.5、0.25 mg·mL^(-1)正丁醇提取液组细胞存活率分别为(103.14±0.14)%、(145.16±0.29)%、(128.45±0.08)%,与模型对照组比较,细胞存活率提高,差异均具有统计学意义。结论刺苋根各不同部位提取物对氯化钙和草酸钠导致的HK-2细胞损伤都有明显的保护作用。OBJECTIVE To explore the preventive effects of the different parts of extract from the root of Amaranthus spinosus L.on Human renal cortical proximal convoluted tubule cells(HK-2)injury induced by calcium oxalate in vitro.METHODS The injury model of HK-2 cells induced by calcium chloride and sodium oxalate was established to screen the active parts of Amaranthus spinosus L.by testing the protective effects of different parts of extracts on HK-2 injury cells through CCK-8 method,and the cells morphology were observed by HE staining.RESULTS Different parts of extracts from the root of Amaranthus spinosus L.improved the survival rate of HK-2 injury cells,which were(105.80±0.08)%、(111.79±0.01)%、(121.43±0.20)%、(128.92±0.00)%by 2,1,0.5,0.25 mg·mL^(-1) ethanol extract,(112.72±0.45)%、(99.94±0.28)%by 0.5,0.25 mg·mL^(-1) petroleum ether extract,(14.04±0.03)%、(111.24±0.08)%、(121.37±0.03)%by 1,0.5,0.25 mg·mL^(-1) ethyl acetate extract,(103.14±0.14)%、(145.16±0.29)%、(128.45±0.08)%by 1,0.5,0.25 mg·mL^(-1) n-butanol extract,all of which showed significant diference compared with the model control group.CONCLUSION The different parts of extracts from the root of Amaranthus spinosus L.have obvious preventive effects on HK-2 cells injury induced by calcium chloride and sodium oxalate.
