黑牛肝菌菌丝体生长培养基的响应面优化及其活力评价

摘  要:本研究通过向PDA培养基中添加不同浓度的无机盐和维生素,以菌丝生长速率为指标,以Box-Behnken响应面法对黑牛肝菌(Phlebopus portentosus)野生株PH-21的生长培养基进行了优化,并对不同培养基上生长的菌丝体中的活性代谢物进行了活性评价。结果表明,培养基中无机盐最佳添加量为FeSO_(4)1.28 mg/mL、CaSO_(4)1.10 mg/mL和MnSO_(4)0.81 mg/mL,维生素最佳添加量为VB_(1)128.5μg/mL、VB_(2)95.1μg/mL和VB_(3)94.8μg/mL,菌丝生长的实际值与预测值相符。优化的培养基中生长获得的菌丝体多糖和三萜类化合物含量分别提高了61.4%和53.5%,1,1-二苯基-2-三硝基苯肼(DPPH)和2,2′-联氮-双-3-乙基苯并噻唑啉-6-磺酸(ABTS)自由基清除率分别提高了89.2%和44.6%,同时可显著减少细胞中丙二醛(malondialdehyde,MDA)含量,缓解超氧化物歧化酶(superoxide dismutase,SOD)、过氧化物歧化酶(peroxidase,POD)和过氧化氢酶(catalase,CAT)活性的衰减。这些结果表明,在黑牛肝菌的培养过程中,可通过向培养基中添加适宜浓度的无机盐和维生素来提高黑牛肝菌的菌丝生长,减缓菌丝老化速度。In this study,the effect of different concentrations of inorganic salts and vitamins on the mycelial growth rate of Phlebopus portentosus wild strain PH-21 was investigated,and then the growth medium was optimized using Box Behnken response surface design,and the activity of bioactive metabolites from mycelial cultures of the strain on different media was evaluated.The results showed that the predicted optimal addition of FeSO_(4),CaSO_(4) and MnSO_(4) were 1.28 mg/mL,1.10 mg/mL and 0.81 mg/mL respectively,and optimum concentration of vitamins were 128.5μg/mL of VB_(1),95.1μg/mL of VB_(2) and 94.8μg/mL of VB_(3).The results of validation experiment were consistent with the predicted values of the model.Moreover,the contents of polysaccharides and triterpenoids extracted from mycelia were increased by 61.4% and 53.5% in the optimized medium compared to the control,respectively.Furthermore,DPPH and ABTS free radical scavenging rates increased by 89.2% and 44.6%,respectively.Meanwhile,the content of malondialdehyde(MDA)in cells was significantly reduced,while alleviate the attenuation of superoxide dismutase(SOD),peroxidase(POD),and catalase(CAT)activities.Comparing with the PDA medium,adding appropriate concentration of inorganic salts and vitamins to the medium could significantly enhanced mycelium growth and delayed hyphal aging.
