摘  要:目的探讨灵芝孢子类产品对模型小鼠肝损伤的改善作用。方法将42只小鼠随机分为空白对照组[等体积0.5%羧甲基纤维素钠(CMC-Na)]、模型组(等体积0.5%CMC-Na)、破壁灵芝孢子粉组(破壁组,0.1 g/kg)、富硒破壁灵芝孢子粉组(富硒破壁组,0.1 g/kg)、灵芝孢子油组(孢子油组,0.1 g/kg)、未破壁灵芝孢子粉组(未破壁组,0.1 g/kg),各7只。尾静脉注射0.25 mg/mL刀豆蛋白A,以复制肝损伤小鼠模型,正常对照组给予等体积生理盐水,隔天1次,连续2周;建模成功后各组小鼠灌胃相应药物或0.5%CMC-Na,每天1次,连续30 d。采用苏木素-伊红(HE)染色,观察小鼠肝组织形态学变化;检测小鼠血清中天门冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)、肿瘤坏死因子-α(TNF-α)、γ干扰素(IFN-γ)及肝组织中超氧化物歧化酶(SOD)、丙二醛(MDA)的水平。结果与模型组比较,破壁组、富硒破壁组、孢子油组小鼠的肝细胞坏死程度均显著减轻,肝索排列较规范,炎性程度减轻;未破壁组小鼠肝细胞坏死程度虽减轻,但坏死处仍有炎性细胞浸润。与模型组比较,破壁组、富硒破壁组、孢子油组、未破壁组小鼠血清中AST,ALT,TNF-α,IFN-γ水平及肝组织中MDA水平均显著降低,肝组织中SOD水平均显著升高(P<0.05)。结论上述4种灵芝孢子类产品具有一定的保肝护肝作用,其机制与修复损伤肝细胞、促进肝细胞生长、抗氧化和免疫调节相关。Objective To investigate the improvement of Ganoderma lucidum spore(GLS)products on liver injury in model mice.Methods Forty-two mice were randomly divided into the blank control group[equal volume of 0.5%sodium carboxymethyl cellulose(CMC-Na)],model group(equal volume of 0.5%CMC-Na),wall-broken GLS powder group(wall-broken group,0.1 g/kg),selenium-enriched wall-broken GLS powder group(selenium-enriched wall-broken group,0.1 g/kg),GLS oil group(broken group,0.1 g/kg),wall-unbroken GLS powder group(wall-unbroken group,0.1 g/kg),seven mice in each group.Mice were injected with 0.25 mg/mL concanavalin A(Con A)via tail vein to duplicate the model mice with liver injury.The mice in the normal control group were given an equal volume of normal saline once every other day for two weeks.After successful modeling,mice in each group were given corresponding drugs or 0.5%CMC-Na by gavage once a day for 30 d.Hematoxylin-eosin(HE)staining was used to observe the morphological changes in liver tissue in mice.The levels of aspartate aminotransferase(AST),alanine aminotransferase(ALT),tumor necrosis factor-α(TNF-α),interferon-γ(IFN-γ)in serum,and the levels of superoxide dismutase(SOD)and malondialdehyde(MDA)in liver tissue were detected.Results Compared with those in the model group,the necrosis degree of liver cells reduced significantly,the arrangement of liver cords was more standardized,and the degree of inflammation reduced in the wall-broken group,selenium-enriched wall-broken group and GLS oil group.Although the necrosis degree of liver cells reduced,there was still inflammatory cell infiltration in the necrotic area in the wall-unbroken group.Compared with those in the model group,the levels of AST,ALT,TNF-αand IFN-γin serum and the level of MDA in liver tissue in the wall-broken group,selenium-enriched wall-broken group,GLS oil group and wall-unbroken group were significantly lower,while the levels of SOD in liver tissue in the above groups were significantly higher(P<0.05).Conclusion The above four GLS products
