摘 要:使用双室微生物电解池(MEC)装置,分别以回流污泥、曝气池污泥、小麦根际土壤为接种源,探究MEC去除模拟废水中氨氮的最佳电压及相关微生物群落变化。结果显示,不同的微生物接种源都有其最佳外加电压,外加电压对氨氮去除率的影响也呈现出不同趋势,以回流污泥为接种源的MEC在0.5 V电压下的氨氮去除率最高为75%。利用高通量测序对微生物群落进行多样性分析,Proteobacteria为硝化反应的优势微生物菌门,在最佳电压下培养前后,以回流污泥和曝气池污泥为接种源的MEC中的Proteobacteria均出现了增长,分别由36.1%、36.4%增加到68.4%、60.7%,而以根际土壤为接种源的MEC中的微生物群落基本未发生明显变化。就微生物组成而言,曝气池污泥和回流污泥的组成较为接近,外加电压对其影响也呈现出一定的相似性,而以根际土壤为接种源的MEC与前两者差别较大。不同接种源微生物的多样性和丰富度均有所不同,结合氨氮去除效果推测,外加电压促使了MEC中硝化功能菌聚集,从而提高了MEC去除氨氮的能力。A two-compartment microbial electrolysis cell(MEC)was used to investigate the optimal voltage and related microbial community changes of ammonia nitrogen removal from simulated wastewater by using reflux sludge,sludge from an aeration tank,and soil from wheat roots as microbial inoculation sources. The results showed that different microbial inoculation sources had their best applied voltage,and the effect of applied voltage on ammonia removal rate also showed different trends. The maximum ammonia removal rate of MEC inoculated with reflux sludge was 75% at 0.5 V.High-throughput sequencing was used to analyze the diversity of microbial communities and Proteobacteria were found to be the dominant microbial phyla for nitrification reaction. Before and after culture,the dominant microflora Proteobacteria in the MEC inoculated with reflux sludge and aeration tank sludge increased from 36.1% and 36.4% to 68.4%and 60.7% under the optimum voltage,respectively,while the microflora in the MEC inoculated with root soil did not change significantly. In terms of microbial composition,the compositions of the aerator tank sludge and reflux sludge were similar,and the influence of applied voltage also showed certain similarity. However,the microbial composition of the reactor inoculated from roots soil was quite different from that of the former two. The diversity and richness of different inoculated microorganisms were different. Combined with the ammonia nitrogen removal effect,it was speculated that the applied voltage promoted the accumulation of nitrifying bacteria in the MEC,thus improving their ammonia nitrogen removal ability.
