摘 要:目的探讨小鼠隐睾模型睾丸组织病理变化与血清抑制素B和抗苗勒管激素水平之间的关系。方法饲养(美国癌症研究所ICR)雄性小鼠50只,利用随机数字表将小鼠随机分为3组:即假手术组、单侧隐睾组和双侧隐睾组。单侧组与双侧组采取手术方式固定对应睾丸至腹腔内,假手术组打开腹腔后睾丸组织未做特殊处理。分别在造模后第10、20、30天3组小鼠分离腹主动脉采血。取双侧睾丸组织清洗后称重,甲醛溶液浸泡保存。用ELISA试剂盒酶联免疫法测量血清两种激素水平,睾丸组织HE染色,光学显微镜下观察依据Johnsen评分标准进行评分。用SPSS 21.0统计分析软件进行统计分析。结果各组实验鼠体质量之间差异均无统计学意义(P<0.05)。睾丸质量、睾丸体积单侧组健侧睾丸与患侧睾丸比较、双侧组与假手术组比较差异均有统计学意义(P<0.05)。血清AMH和INH-B浓度3组间比较差异均有统计学意义(P<0.05),其中假手术组血清两种激素水平最高[AMH(12.79±7.99)pmol/L,INH-B(29.25±11.56)ng/L)],单侧组次之[AMH(8.79±3.08)pmol/L,INH-B(22.56±13.68)ng/L)],双侧组最低[AMH(5.53±3.30 pmol/L,INH-B(12.64±8.07ng/L)]。睾丸组织Johnsen评分组间比较,单侧组患侧睾丸评分(5.40±0.62分)明显低于健侧(8.40±0.42分),双侧组评分[(4.60±0.55)分]明显低于假手术组[(9.80±0.61)分],差异均有统计学意义(P<0.05)。结论隐睾症可引起小鼠睾丸组织功能障碍,3组小鼠血清INH-B和AMH激素水平数据对比,双侧组两种激素水平最低,假手术组水平最高。这一差异性跟睾丸参数及病理评分完全符合。可以认为血清INH-B和AMH是一种简便、准确的血清标志物,可以很好地评估隐睾睾丸组织的功能。Objective To explore the relationship between testicular histopathological changes and serum inhibin-B(IHN-B)and anti-Mullerian hormone(AMH)levels in mouse cryptorchidism model.Methods A total of 50 ICR male mice were randomly divided into three groups:sham operation group,unilateral cryptorchidism group and bilateral cryptorchidism group.The unilateral and bilateral groups fixed the testis to the abdominal cavity by operation,and the sham operation group just opened the abdominal cavity.Blood was sampled from abdominal aorta on days 10,20 and 30 after modeling.Testicular tissues from both sides were harvested,cleaned and weighed,then soaked in formaldehyde solution for preservation.The serum levels of IHN-B and AMH were measured with ELISA.After HE staining of testicular tissues,the histopathological changes-were observed with an optical microscope according to Johnsen scoring criteria.SPSS 21.0 software was used for statistical analysis.Results There was no significant difference in body weight among the three groups(P<0.05).There were significant differences between healthy testis and affected testis in the unilateral group,and mean testicular volume(weight)between the bilateral group and sham operation group(P<0.05).There were significant differences in serum AMH and INH-B among the three groups(P<0.05).The sham group had the highest serum levels[AMH(12.79±7.99)pmol/L,INH-B(29.25±11.56)ng/L],followed by the unilateral group[AMH(8.79±3.08)pmol/L,INH-B(22.56±13.68)ng/L]and the bilateral group[AMH(5.53±3.30)pmol/L,INH-B(12.64±8.07)ng/L].Johnsen score of the affected testis(5.40±0.62)was significantly lower than that of the healthy testis in the unilateral group(8.40±0.42),and lower in the bilateral group(4.60±0.55)than in the sham operation group(9.80±0.61)(P<0.05).Conclusion Cryptorchidism can cause testicular tissue dysfunction in mice.The serum INH-B and AMH levels were the lowest in the bilateral group and the highest in the sham operation group,which is consistent with testicular parameters and pa
