摘 要:芹菜种子小,且含有大量的萌发抑制物质,给其种苗繁育带来困难。采用培养皿培养和穴盘培养两种方式,用不同浓度(0、5%、10%、20%、40%)NaClO处理‘六合黄心芹’和‘赛雪’芹菜种子,对种子萌发指标、幼苗生长指标及光合参数进行测定,并分析幼苗中叶绿素含量变化以及相关基因表达。结果表明:5%次氯酸钠溶液处理芹菜种子,其发芽率、发芽势、发芽指数和活力指数最高,幼苗生长势最强,幼苗的壮苗指标、叶绿素含量以及净光合速率也有一定程度提高,相关基因的相对表达量也较高。这些结果说明次氯酸钠处理促进芹菜种子的适宜浓度为5%。Celery seeds are small and contain a large amount of germination-inhibiting substances,which brings difficulties to celery seeding propagation. The seeds of‘Liuhe Huangxinqin’and‘Saixue’were treated with different concentrations(0,5%,10%,20%,40%)of NaClO cultured in petri dish and acupoint dish. The seed germination index,seedling growth index and photosynthetic parameters were measured. In addition,the changes of chlorophyll content and related gene expression in seedlings were analyzed. The results showed that celery seeds treated with 5% NaClO had the highest germination rate,germination potential,germination index and vigor index,and the strongest seedling growth potential. And the seedling vigor index,chlorophyll content and net photosynthetic rate were also increased to some extent. Also,the relative expression levels of related genes were also higher. These results suggested that the appropriate concentration for NaClO to promote celery seeds is 5%.
