摘 要:目的:探讨广西毛冬青根对放射脑损伤小鼠认知功能障碍的改善作用及其机制。方法:将100只SPF级雄性昆明小鼠随机分为对照组、毛冬青组、放射组和放射+毛冬青组,每组25只。毛冬青组和放射+毛冬青组小鼠于放射前给予毛冬青溶液连续灌胃7 d(1 mL/30 mg),其余各组给予等量生理盐水。放射组和放射+毛冬青组采用^(60)Coγ射线照射小鼠脑部建立认知障碍模型,放射前连续给药7 d;对照组和毛冬青组置于同等条件中,照射剂量为0 Gy。通过神经行为、ELISA、HE染色以及免疫荧光等方法观察广西毛冬青根干预对小鼠空间记忆功能、海马区组织形态结构以及小胶质细胞、星形胶质细胞活性的影响。结果:与对照组相比,放射组小鼠出现记忆功能障碍,海马区神经细胞核固缩,神经胶质细胞激活,IL-6、Iba-1、GFAP水平提高(P<0.05);与放射组相比,放射+毛冬青组小鼠记忆能力、海马区细胞的形态结构得到改善,IL-6、Iba-1和GFAP水平下降(P<0.05)。结论:广西毛冬青根可以改善放射性脑损伤小鼠的认知功能,其机制可能与抑制神经胶质细胞活化有关。Objective:To investigate the ameliorative effect of Ilex pubescens var.kwangsiensis roots on cognitive impairment in mice with radiation-induced brain injury and its mechanism.Methods:One hundred male SPF-grade Kunming mice were randomly divided into control group,Ilex pubescens group,radiation group and radiation+Ilex pubescens group,with 25 mice in each group.Mice in the Ilex pubescens and radiation+Ilex pubescens groups were given Ilex pubescens solution by continuous gavage for 7 d(1 mL/30 mg)before radiation,and the rest groups were given equal amount of saline.The mice in the radiation and radiation+Ilex pubescens groups were irradiated with^(60)Coγ-rays to establish a cognitive impairment model,and the control group and the Ilex pubescens group were placed in the same conditions with an irradiation dose of 0 Gy.The effects of Ilex pubescens var.kwangsiensis root intervention on the spatial memory function,the morphological structure of the hippocampal region and the activities of microglia and astrocytes were observed by neurobehavioral test,ELISA,HE staining and immunofluorescence assay.Results:Compared with the control group,mice in the radiation group showed memory dysfunction,pyknosis in the hippocampus,glial cell activation,and increased levels of IL-6,Iba-1,and GFAP(P<0.05);compared with the radiation group,mice in the radiation+Ilex pubescens group showed improved memory capacity and morphological structure of cells in the hippocampus,and decreased levels of IL-6,Iba-1,and GFAP(P<0.05).Conclusion:Ilex pubescens var.kwangsiensis roots can improve the cognitive function of mice with radiation-induced brain injury,and the mechanism may be related to the inhibition of glial cell activation.
