摘 要:目的:探讨厌氧环境对铜绿假单胞菌(P.a)生长、生物被膜形成、毒力因子产生及群体感应系统(QS)的影响。方法:复苏P.a野生菌株PAO1和两种QS基因缺陷型菌株PAO1 lasIrhlI、PAO1 lasRrhlR,并在有氧和厌氧环境下通过生长试验、生物被膜形成试验、毒力因子测定来检测氧环境差异及QS对P.a的影响。结果:有氧条件下,PAO1、PAO1 lasIrhlI、PAO1 lasRrhlR生长能力无明显差异,PAO1生物被膜形成能力及毒力因子水平高于PAO1 lasIrhlI、PAO1 lasRrhlR(P<0.05)。相较有氧条件,PAO1菌株在厌氧条件下生长减缓,生物被膜形成减少,毒力因子水平下降(P<0.05);而厌氧条件下PAO1 lasIrhlI和PAO1lasRrhlR生长减缓,生物被膜形成能力及毒力因子水平与有氧条件下比较无明显差异;厌氧条件下,3种菌株生长、生物被膜形成及毒力因子水平比较,差异无统计学意义(P>0.05)。结论:P.a在厌氧环境下生长能力、生物被膜形成能力和毒力因子活性均减弱,QS系统对厌氧条件下P.a的生物被膜形成和毒力因子的调节作用受到抑制。Objective:To investigate the effects of anaerobic conditions and quorum sensing(QS)on the growth,biofilm formation,virulence factor production and of Pseudomonas aeruginosa(P.a).Methods:P.a wild strain PAO1 and two QS gene-deficient strains PAO1 lasIrhlI and PAO1 lasRrhlR were resuscitated,and the effects of oxygen conditions and QS on P.a were examined by growth assay,biofilm formation assay,and virulence factor assay under aerobic and anaerobic conditions.Results:There was no significant difference in the growth ability of PAO1,PAO1 lasIrhlI,and PAO1 lasRrhlR under aerobic conditions,and the biofilm formation ability and virulence factor levels of PAO1 were higher than those of PAO1 lasIrhlI and PAO1 lasRrhlR(P<0.05).Compared with aerobic conditions,the growth of PAO1 strains under anaerobic conditions was slowed down,biofilm formation was reduced and virulence factor levels decreased(P<0.05);while the growth of PAO1 lasIrhlI and PAO1lasRrhlR under anaerobic conditions was slowed down,biofilm formation ability and virulence factor levels were not significantly different from those under aerobic conditions;under anaerobic conditions,the growth,biofilm formation and virulence factor levels of the three strains were not statistically different(P>0.05).Conclusion:The growth ability,biofilm formation ability and virulence factor activity of P.a were reduced under anaerobic conditions,and the regulation of biofilm formation and virulence factor of P.a by QS system was inhibited under anaerobic conditions.
