摘  要:目的考察谷氨酰胺对骨肽生物活性测定方法——UMR106细胞增殖试验的影响。方法使用液质联用测定骨肽原液中谷氨酰胺含量,采用CCK-8法检测谷氨酰胺对UMR106细胞增殖影响的浓度范围,以及骨肽原液对UMR106细胞的增殖作用。同时对骨肽原液浓度与相应谷氨酰胺浓度,以及谷氨酰胺试验应用浓度(骨肽原液浓度1 mg/mL)与相应细胞增殖率进行相关性分析。结果骨肽原液中谷氨酰胺含量差异较大,且UMR106细胞对谷氨酰胺较为敏感。骨肽原液浓度与其相应谷氨酰胺含量、谷氨酰胺试验应用浓度与相应细胞增殖率均无显著相关性(r分别为0.1463和0.5471,P均>0.05)。结论骨肽原液中谷氨酰胺不会对骨肽生物活性测定方法——UMR106细胞增殖试验产生影响。Objective To investigate the effect of glutamine on UMR106 cell proliferation test,a method for determination of biological activity of osteopeptide.Methods The glutamine content in bulk of osteopeptide was determined by liquid chromatography-mass spectrometry(LC-MS).The concentration range of glutamine influencing the proliferation of UMR106 cells and the effect of bulk of osteopeptide on the proliferation were evaluated by CCK-8 method.Meanwhile,the correlation of osteopeptide concentration with the corresponding glutamine concentration,and that of the working concentration of glutamine(the osteopeptide concentration was 1 mg/mL)with the corresponding cell proliferation rate were analyzed.Results The glutamine contents varied greatly in bulks of osteopeptide from various manufacturers.UMR106 cells were relatively sensitive to glutamine.There was no significant correlation between the osteopeptide concentration and the corresponding glutamine content,and between the working concentration of glutamine and the corresponding cell proliferation rate(r=0.1463,0.5471,each P>0.05).Conclusion Glutamine in bulk of osteopeptide showed no effect on UMR106 cell proliferation test as a method for determination of biological activity of osteopeptide.
